Cholesterol esterase is a hydrolase that catalyzes the conversion of ingested cholesterol ester to free cholesterol and free fatty acid [1]. Cholesterol esterase exhibits proatherogenic properties by facilitating the conversion of the larger and less atherogenic lipoprotein particles to smaller and more atherogenic lipoprotein subspecies [2]. Cholesterol esterase can be covalently immobilized on the polyaniline film to develop a biosensor for cholesterol measurement[3].
In vitro, Cholesterol esterase (100mU/ml) treatment for 3 hours promotes the proliferation and migration of human umbilical vein endothelial cells (HUVECs) [4].

Preparation Method | HUVECs were incubated with EBM-2 medium supplemented with 2% fetal calf serum (FCS) in a humidified atmosphere at 37°C with 5% CO2. HUVEC migration was determined using a 24-well chemotaxicell chamber with polycarbonate filter (8-μm pore size). HUVECs (2×104 cells) in serum-free medium (100μl) were applied in the upper compartment of the chamber. The medium containing Cholesterol esterase (100mU/ml) was added to the lower compartment of the chamber and incubated (37°C; 3 hours). After removing Cholesterol esterase and a further incubation (37°C; 24 hours), the non-invading cells on the upper surface of the filter were completely wiped off with a cotton swab and filters were fixed (70% ethanol in PBS) and stained (hematoxylin). The number of migrated cells was counted in 10 random fields at 200× magnification under the microscope. |
