DC661 est un composé de petite molécule capable d'inhiber la palmitoyle-protéine thioestérase 1 (PPT1) et de supprimer efficacement l'autophagie cellulaire, agissant comme agent anti-lysosomal[1-2]. DC661 est principalement utilisé dans la recherche liée au cancer[3-4].
Le pré-traitement in vitro avec DC661 (1.5–3μM) aux cellules endothéliales de la veine ombilicale humaine (HUVEC) pendant 30 minutes, suivi d'une exposition à l'hypoxie pendant 5,5 heures, a significativement inhibé l'expression de PPT1 et réduit les réponses angiogéniques[5]. Le pré-traitement avec DC661 (0.5–3μM) aux cellules de carcinome hépatocellulaire Hep3B et Hep1-6 pendant 6-24 heures, suivi d'une stimulation avec le sorafénib (1.5–10μM) pendant 24–48 heures a significativement supprimé l'expression de PPT1 et l'activité autophagique, tout en augmentant la sensibilité au sorafénib[6].
In vivo, l'injection intra-péritonéale de DC661 (3mg/kg/jour) pendant 21 jours chez les rats porteurs de tumeurs de mélanome (rats avec invalidation conditionnelle de Ppt1) n'avait aucune effet sur l'initiation ou la croissance des tumeurs de mélanome[7]. Dans un modèle de rats Wistar exposés au plomb (200mg/kg/jour pendant 6 semaines), l'administration stéréotaxique de DC661 (10mM, 5μl/jour pendant 6 semaines) dans l'hippocampe a considérablement inhibé le niveau d'autophagie dans les neurones hippocampiques et augmenté la régulation de l'activité de la voie de signalisation IGF-1. Le traitement avec DC661 a significativement activé la voie IGF-1/PI3K/Akt/mTOR, inversé l'autophagie excessive induite par le plomb et amélioré la fonction neuronale hippocampique[8].
