Nusinersen est un SMN-ASO qui augmente le niveau de SMN au moyen de bloquer un silencieux d'épissage intronique dans SMN2, ainsi donc, facilite l'inclusion de l'exon 7 et la génération de transcrits FL-SMN2[1].
Nusinersen(6.5mg/kg; I.C.V.) restaure robustement la courbe de croissance des rats SMN∆7, améliore la fonction motrice et réduit dramatiquement la perte de αMN et prévient la perturbation et la perte des corps de Cajal canoniques (CBs) dans la moelle épinière des rats SMA. D'ailleur, le traitement avec Nusinersen normalise la distribution de la machinerie de synthèse protéique et des mRNAs polyadénylés chez les rats SMNΔ7[2]. Nusinersen a été administré aux lapins par voie intrathécale (IT) avec une dose bolus de 1.8 mg/kg pour le premier groupe et par voie intraveineuse (IV) avec une dose bolus de 1 mg/kg pour le deuxième groupe, respectivement. Cmax, AUClast et C0 d' IV étaient 15.6±4.88μg/mL, 17.8±4.10 h*μg/mL et 17.6±6.20μg/mL; Tmax, Cmax et AUClast d' IT étaient de 1h, 6.0±0.10μg/mL et 12.4±1.15 h*μg/mL, respectivement. La biodisponibilité calculée de Nusinersen était 33 %[3].

Méthode de préparation | Pour rechercher les effets du traitement avec nusinersen chez αMNs des rats SMN∆7, nous avons utilisé 32 animaux (16 rats WT et 16 rats SMN∆7), qui ont été divisés en quatre groupes. Les rats WT et les rats SMN∆7 ont été administrés respectivement par 10µl de nusinersen à la dose de 6.5mg/kg (groupe de nusinersen) ou par 10µl d'une saline à 0.9% (groupe véhicule). Nusinersen ou le véhicule a été injecté le jour de la naissance (P0) par voie intracérébroventriculaire (ICV) à l'aide d'une seringue de 10µl et d'une aiguille personnalisée de gauge 33. |
