Description of Thalidomide

Thalidomide est un sédatif non addictif et non barbiturique qui inhibe le cereblon, avec des propriétés anti-inflammatoires et anti-angiogéniques[1]. Thalidomide se fixe aux sites promoteurs riches en GC par intercalation et réduit ainsi la transcription d'IGF-1 et de FGF-2 et inhibe l'angiogenèse pour excercer les effets tératogènes [2]. Thalidomide a été utilisée pour atténuer l'inflammation neuronale et bloquer la vascularisation excessive, le développement et les métastases tumorales[3].

In vitro, un traitement par Thalidomide durant 72 heures a inhibé la prolifération des cellules MG-63 et des cellules U2OS, avec des valeurs d' IC50 de 94.76µg/ml et 156.61µg/ml, respectivement[4]. Un traitement par Thalidomide (150µg/ml) durant 12 heures a inhibé l'assemblage de structures tubulaires dans les cellules ECV 304 et a induit une réorganisation du cytosquelette[5]. Un prétraitement par Thalidomide (1mM) durant 30 minutes inhibe l'activation transcriptionnelle de NFκB induite par IL-1β dans les cellules Caco-2 et la production d'IL-8, et a inhibé la translocation nucléaire de NFκB et la dégradation de IκB[6].

In vivo, un traitement par Thalidomide par administration orale à une dose de 200mg/kg/jour durant 30 jours a inhibé le développement tumoral et les métastases de xénogreffe A549 chez la souris [7].

Des cellules MG-63

Méthode de préparation

Des cellules MG-63 ont été cultivées dans du milieu de Eagle modifié par Dulbecco (DMEM) avec 10% (v/v) de sérum de veau foetal (SVF), 100μg/ml de streptomycine et 100U/ml de pénicilline à 37°C dans un atmosphère à 5% de CO2. Des cellules MG-63 (1×104) ont été déposées dans chaque puits d'une plaque de 96 puits durant une nuit avant le traitement par Thalidomide. Thalidomide a été ajoutée aux cellules à diverses concentrations (0, 12.5, 25, 50, 100, 200 et 400µg/ml), et les cellules ont été incubées à 37°C dans un incubateur à 5% de CO2 durant 72h. La viabilité cellulaire a été analysée.

Modèles animaux

Des souris nues BALB/c

Méthode de préparation

Des souris nues BALB/c femelles ont été hébergées dans un installation spécialisée de soins aux animaux, dans un pièce à température constante (25°C), avec contrôle de l'humidité, et un cycle lumière/obscurité de 12/12h, avec un accès libre à la nourriture et à l'eau. Des cellules A549 ont été remises en suspension dans PBS. Ensuite, 1×107 cellules dans 0.1ml de PBS ont été injectées dans la veine latérale de la queue de souris nues âgées de 6 semaines. Deux semaines après l'administration, des tumeurs de 100 à 200mm3 étaient apparentes chez toutes les souris. Les souris ont été divisées en deux groupes (n=6) et ont reçu par voie orale soit un véhicule contrôle (huile de soja), soit Thalidomide (200mg/kg/jour) en suspension dans l'huile de soja. Les souris ont été traitées chaque jour durant 30 jours avec thalidomide ou le véhicule contrôle, et la croissance tumorale a été mesurée deux fois par semaine avec un pied à coulisse.
