Tarlatamab is a half-life extended bispecific T-cell engager molecule targeting delta-like ligand 3 (DLL3) [1]. Tarlatamab binds to DLL3 on the surface of tumour cells and CD3 on the surface of cytotoxic T lymphocytes (CTLs), resulting in T-cell activation, release of inflammatory cytokines, and CTL-mediated cell death of DLL3-expressing tumor cells [2]. Tarlatamab has been widely used to enhance T-cell infiltration and anti-tumor responses, as well as to induce the regression of xenograft tumors[3].
In vitro, Tarlatamab treatment (0.1nM) for 48 hours significantly inhibited the viability of SHP-77 cells and induced the production of cytokines and the release of cytotoxic granules[4].
In vivo, Tarlatamab treatment via intraperitoneal injection at a dose of 3mg/kg, once a week for 3 weeks, significantly inhibited tumor growth in the mice with small cell lung cancer (SCLC) and activated the activity of infiltrating T cells in the tumor tissues[4].

Preparation Method | SHP-77 cells were cultured in RPMI-1640 medium supplemented with 10% fetal bovine serum, 100U/ml penicillin, and 100μg/ml streptomycin at 37°C with 5% CO2 and 95% saturated atmospheric humidity. SHP-77 cells were co-cultivated with purified human CD3+ T cells at a cell ratio of 5:1 with a dose range of Tarlatamab (0, 0.001, 0.01, 0.1, 1, and 10nM) for 48 hours, cell viability was analyzed. |

Preparation Method | Female NOG mice were maintained under a 12h light and 12h dark cycle at a temperature of 25°C and humidity of 50%±10%, free with water and diet. After one week of adaptation, mice were implanted subcutaneously with patient-derived SCLC tumor fragments. When the average tumor volume was 94-123mm3, mice were administered 3mg/kg Tarlatamab by intraperitoneal injection on days 1, 8, and 15. Tumor volumes and body weights were measured twice a week until study end at day 23. |
