Description of UDP-α-D-Glucose (sodium salt)

UDP-α-D-Glucose (sodium salt) is an endogenous nucleotide sugar composed of uridine diphosphate and α-D-glucose. UDP-α-D-Glucose has a well established biochemical role as a glycosyl donor in the enzymatic biosynthesis of carbohydrates. UDP-α-D-Glucose is also an agonist of the G protein-coupled receptor KIAA0001 (P2Y14), which is expressed in neutrophils and is involved in activation of inflammatory signaling[1][2].

In vitro, UDP-α-D-glucose (100μM; 4h) increased ERK1/2 phosphorylation and upregulated IL-8 and CCL2 mRNA expression in MDCK-C11 cells[3]. UDP-α-D-glucose (30μM; 2h) enhanced the random migration of eosinophils[4].

In vivo, UDP-α-D-Glucose (200mg/kg/d; s.c.; 6d) promoted the mobilization of hematopoietic stem and progenitor cells (HSPCs), as evidenced by a significant increase in circulating Colony-Forming Units in Culture (CFU-Cs) in the blood of both B6 and BALB/c mice, including CFU-Granulocyte, Macrophage (CFU-GM), Burst-Forming Unit-Erythroid (BFU-E), and CFU-Granulocyte, Erythroid, Macrophage, Megakaryocyte (CFU-GEMM)[5]. UDP-α-D-Glucose (500-2000mg/kg; i.p.; once) dose-dependently reduced gastric emptying in the conscious rat[6].

Preparation Method

Neutrophils (2×106/ml) were incubated in HBSS in the presence of 100μM UDP-α-D-Glucose or 100μM ATP for 60min. At the times indicated, cells were centrifuged and nucleotides present in the supernatant were separated by HPLC (Waters) via a Nova-Pack C18 column. Absorbance at λ=260nm and radioactivity were quantified on-line as previously described.

Applications

Incubation of neutrophils with 100μM UDP-α-D-Glucose for 60min resulted in negligible metabolism of the nucleotide-sugar, i.e., <1% of the initial UDP-α-D-Glucose was recovered as UMP. In contrast, neutrophils rapidly hydrolyzed 100μM ATP, displaying an apparent hydrolysis rate of Vhyd=7.3nmol/min×106 cells.

Preparation Method

Male CD rats were acclimatized to being singly housed in cages for at least 3 days before experimental use. The animals were fasted overnight before the experiment but were allowed free access to drinking water at all times. On the day of the experiment, UDP-α-D-Glucose dissolved in 0.9% wt/vol sodium chloride was administered by intraperitoneal (i.p.) injection at 500, 1000, and 2000mg/kg in a total volume of 2ml/kg. Control animals received 0.9% wt/vol i.p. sodium chloride (saline), and, 10min later, 3ml of the nutrient meal was administered orally. 60min later, animals were culled, stomach contents were weighed, and the percentage of gastric emptying was calculated.
