Melatonin은 소뇌상체에서 생성되는 호르몬으로 Melatonin 수용체를 활성화할 수 있다. Melatonin은 수면에 작용하고 중요한 항산화 및 항염증 특성을 가지고 있다[1][2][3]. Melatonin은 새로운 선택적 ATF-6 억제제이고 COX-2 저하를 통해 인간 간암 세포 사멸을 유도한다[4].
Melatonin은 동물에서 활성화된 PTEN, RSK-1, mTOR 및 AMPKα 키나아제 수준을 증가시키고 ERK-1/2 인산화와 Bad 인산화를 약하게 억제하고 S6 표준의 단백질, 4E-BP1, GSK-3α 및 GSK-3β의 인산화를 현저하게 억제하고 PRAS40 인산화를 약간 증가시킨다[1]. Melatonin은 대뇌 피질에서 Aβ1-42가 유도하는 신경독성 및 아스트로사이트 활성을 개선한다. Melatonin은 또한 Aβ1-42가 유도하는 Reelin과 Dab1 발현의 감소를 차단한다[2]. Melatonin 처리와 NLRP3-/- 결핍은 쥐에서 NF-κB 및 NLRP3 신호전달 경로를 현저하게 억제한다. Melatonin 처리와 NLRP3-/- 결핍은 시계 유전자 발현 패턴을 공유하고 쥐 심근세포의 형태를 개선한다[3].

동물 실험 | 21-26 g의 체중을 가진 성체 수컷 C57BL/6j 쥐 두 그룹을 네 그룹 중 하나로 무작위로 배정해서 (i) 대차물질 (50 µL isotonic saline /5% ethanol), (ii) ethanol (4 mg/kg, 0.9% isotonic saline /5% ethanol에 녹인), (iii) Wortmanni 그리고 (iv) melatonin/Wortmannin을 재관류 후 즉시 복강내 투여하였습니다. 첫 번째 그룹에서는 쥐를 30분 동안의 초점성 뇌허혈 (FCI) 과 72시간 재관류에 노출시켜 문상체의 분산된 허혈 손상을 평가하고 신호전달 경로를 분석하였습니다 (그룹당 n=7). 두 번째 쥐 그룹은 90분 동안의 FCI와 24시간 재관류에 노출되어 뇌졸중 발달, 뇌 부종 및 IgG 유출을 분석하였습니다 (그룹당 n=7). |
