Amarogentin es un glucósido secoido natural que se extrae de las raíces de plantas como Swertia y Gentiana. Pues cuneta con múltiples acciones inmunomoduladores, antidiabéticos y antitumorales[1-2]. Y tiene gran potencial en estudios de cáncer gástrico, colorrectal, diabetes y también neuroprotección[3-4].
En cultivos celulares, se pretrataron células PC12 con Amarogentin (1-10µM) durante 24 horas. Luego se expusieron al estrés oxidativo provocado por peróxido de hidrógeno (0.9mM). Amarogentin frenó mucho el aumento de especies reactivas de oxígeno y malondialdehído. Subió la expresión génica de SOD2, CAT, GPx, Nrf2 y Bcl-x1. Y mejoró la actividad enzimática de SOD2, logrando así proteger las neuronas[5]. Se incubaron células HepG2 y Huh7 (tratadas con ablación por radiofrecuencia incompleta) con Amarogentin (120μg/ml) durante 24 horas. El compuesto redujo la proporción de células CD133 positivas y la secreción de VEGFA. Bloqueó la vía VEGFA/Dll4/Notch1 por un mecanismo dependiente de p53. Y con eso frenó la angiogénesis[6].
En animales, se administró Amarogentin por vía oral (50 o 100mg/kg/día) durante 5 semanas a ratas con osteoporosis por deficiencia de estrógenos. Aumentó mucho la densidad mineral ósea de todo el cuerpo y de la columna lumbar. Mejoró los marcadores séricos de formación ósea, osteocalcina, telopéptido C del colágeno tipo I, pro-péptido N del procollágeno tipo I y fosfatasa alcalina ósea. Rejó las citocinas inflamatorias IL-1β, IL-17 y TNF-α. También estimuló la diferenciación de los osteoblastos regulando la vía Nrf-2/MAPK/ERK[7]. Se inyectó Amarogentin por vía intravenosa (0.5mg/kg/día) durante una semana a ratas con diabetes tipo 1 inducida por estreptozotocina. Disminuyó la glucosa en ayunas. La expresión aumentó de GLUT4 en músculo esquelético. E inhibió la actividad de la PEPCK en el hígado[8].
