Description of Atosiban

Atosiban est un antagoniste mixte des récepteurs de l'ocytocine et de la vasopressine. Il inhibe efficacement les contractions utérines et prolonge la grossesse en bloquant de manière cmpétitive les récepteurs de l'ocytocine sur le muscle lisse utérin[1, 2]. Atosiban est un agent tocolytique peptidique synthétique utilisé dans le traitement du travail prématuré[3].

In vitro, le traitement de cellules de myomètre humain avec Atosiban(10μM) a significativement réduit l'activation intracellulaire de la kinase p38 induite par l'ocytocine (OT) et la régulation à la hausse de la COX-2[4]. Le traitement de cellules endothéliales de veine ombilicale humaine (HUVEC) avec Atosiban (10μM) a significativement inhibé l'activité du promoteur du facteur inductible pa l'hypoxie -1 α (HIF-1α) et a réduit l'expression des ARNm et des protéines de HIF-1α dans ces cellules [5].

In vivo, chez des rats modèles d'endométriose expérimentale, un traitement par injection intrapérionéale d'Atosiban (0.5mg/kg/jour)pendant 21 jours a significativement réduit le volume des implants endométriotiques et a considérablement diminué le niveau d'expression de l'antigène nucléaire de prolifération cellulaire (PCNA) dans les tissus implantés [6]. Chez des rats gestantes, un traitement par injection intrapéritonéale d'Atosiban (6mg/kg/jour) pendant 21 jours a significativement augmenté le niveau de stress oxydatif dans le plasma et le tissu cardiaque des ratons nouveau-nés[7].

Cellules musculaires lisses myométriales prénatales

Méthode de préparation

Des cellules musculaire lisses myométriales prénatales ont été stimulées par de l'ocytocine (OT) à 10 nM, en présence ou en l'absence d'Atosiban (10μM) pendant 5min, 15min, 30min, 2h, 4h et 6h. L'expression de la sous-né p65 de NF-κB phosphorylé, des protéines ERK1/2, de p38 MAPK, ainsi que celle de COX-2 et de p-cPLA2, a été détectée par Western blot.

Méthode de préparation

Une endométriose a été induite chirurgicalement chez 35 rates Wistar durant la phase d'oestrus. Quatre semaines après cette procédure, une relaparotomie a été pratiquée. La viabilité et les dimensions des foyers d'endométriose ont été enregistrées. Les rates ont ensuite été réparties aléatoirement en trois groupes. Dans le premier groupe (n=8), une dose quotidienne de 0.2mL de NaCl à 0.9% a été injectée par voie intrapéritonéale (i.p.) (groupe témoin). Dans les deuxième et troisième groupes (n=8 et n=8), d' Atosiban à 0.5mg/kg/jour i.p.1 et du diltiazem à mg/jour i.p. ont été administrés respectivement. Le traitement a été poursuivi pendant 21 jours consécutifs, approximativement à la même heure chaque jour. A la fin du traitement, une laparotomie a été réalisée et les dimensions des foyers d'endométriose ont été de nouveau mesurées. Les implants endométriaux ont été prélevés pour des études histologiques et immunohistochimiques. Les volumes des implants endométriotiques ont été mesurés et des analyses immunohistochimiques ont été effectuées, puis comparés entre les groupes.

Domaines d'application

A la suite du traitement par Atosiban, les volumes des implants endométriotiques ont diminué de manière significative. Les taux d'expression de l'antigène nucléaire de prolifération cellulaire (PCNA) ont été significativement réduits dans les groupes Atosiban et diltiazem par rapport au groupe témoin.
