GGsTop est un inhibiteur irréversible de γ-glutamyl transférase (GGT) (Ki = 170μM).[1]. GGsTop se lie de manière covalente au site actif de GGT, inhibant son activité catalytique, régulant ainsi l'état réducteur intracellulaire, affectant le métabolisme des acides aminés et modulant l'équilibre redox cellulaire[2-3]. GGsTop est principalement utilisé pour atténuer les dommages hépatiques [4].
Dans les cellules humaines du ligament parodontal (hPLCs), la prolifération de hPLCs a été stimulée après un traitement avec GGsTOP (0-50μM; 7j) [5]. Dans les cellules HCC-1806, l'activité du glutathione a été inhibée après un traitement avec GGsTOP (0.01-10000nM; 4h) [6].
Dans un modèle murin de tumeur avec neutropénie induite par cyclophosphamide, GGsTop (5mg/kg; ip; 6j) supprime l'effet promoteur de croissance tumoral du facteur de stimulation des colonies de granulocytes [7]. Chez les rats Otsuka Long-Evans Tokushima Fatty (OLETF), GGsTop (1mg/kg; iv; injection unique) a entraîné une réduction significative des taux sériques d'ALT, d'AST et de γ-GT [8].

Méthode de préparation | La viabilité cellulaire a été mesurée dans les cellules humaines du ligament parodontal (hPLCs) en utilisant le test au bromure de 3-(4,5-diméthylthiazol-2-yl)-2,5-diphényltétrazolium (MTT). Brièvement, hPLCs (2.5×103cellules/puits) ont été ensemencées dans des plaques de 96 puits et laissées s'adhérer et croître pendant 24h. Les cellules ont été mises en carence toute une nuit, puis incubées avec 0, 5, 10, 20 ou 50μM de GGsTOP pendant 1, 3, 5 et 7 jours. Le milieu était remplacé tous les deux jours. Ensuite, 20μl de MTT ont été ajoutés à chaque puits au moment indiqué et les cellules ont été incubées pendant 4h. Le milieu a ensuite été retiré et 150μl de diméthyl sulfoxide (DMSO) ont été ajoutés à chaque puits pendant 10min. |

Modèles animaux | Modèle murin de tumeur avec neutropénie induite par le cyclophosphamide |
Méthode de préparation | Des cellules de lymphome EL4 cells (4 × 105 cellules/souris) ont été injectées par voie sous-cutanée dans le flanc inférieur droit de souris C57BL/6J. Sept jours après l' inoculation des cellules EL4, les souris ont reçu une dose unique de cyclophosphamide (CPA,100mg/kg) par administration intrapéritonéale pour créer un modèle partir du même jour, les souris expérimentales ont reçu des injections intrapértonéales de PBS, de GGsTop (5mg/kg) et de facteur de stimulation des colonies de granulocytes (G-CSF, 200μg/kg) recombinant pendant six jours consécutifs. Avant l'administration des agents mentionnés, du sang rétro-orbitaire(environ 75μL) était prélevé quotidiennement pour une analyse par cytométrie en flux et la détermination de la numération globulaire à l'aide d'un analyse hématologique automatisé Sysmex XT-2000i. |
