MPP dihydrochloride est un antagoniste sélectif du récepteur à l'estrogène α (ERα) [1]. MPP dihydrochloride bloque la signalisation des estrogèènes, induisant ainsi l'apoptose cellulaire[2]. MPP dihydrochloride est utilisé pour traiter le cancer [3].
Dans les cellules MCF-7, un traitement avec MPP dihydrochloride (5μM, 15μM, 20μM; 1h) a amélioré de manière dose-dépendante l'inhibition de la croissance cellulaire silibinine-induite[4]. Dans les cellules SKOV3, le traitement avec MPP dihydrochloride (20μM; 24h) a significativement augmenté le nombre de cellules [5]. Dans les cellules SKOV3 et OV2008, le traitement avec MPP dihydrochloride (10pM, 100pM, 1000pM; 24h) a considérablement inhibé la croissance cellulaire dans les deux lignées cellulaires [6]. Dans les cellules stromales endothéliales ectopiques (eESCs), un traitement avec MPP dihydrochloride (2.7nM; 48h) augmente le niveau d'autophagie dans eESCs [7].
Dans le modèle de rats portant une tumeur sous-cutanée de cellules SKOV3, des traitements avec MPP dihydrochloride (1mg/kg; ip; 35j) réduisent la croissance des cellules tumorales in vivo chez les rats[6]. Dans les rats SERT+, MPP dihydrochloride (2mg/kg; sc; 14j) atténue le développement de l'hypertension pulmonaire (HP) chez les rats féminins SERT+ [8]. Dans les rats C57/BL6, le traitement avec MPP dihydrochloride (200μg/kg; ip; 7j) a significativement réduit l'expression de rictor, p-AKT(Ser473)/AKT, p-cofilin (Ser3), et profilin-1 [9].

Méthode de préparation | Les cellules MCF-7 ont été ensemencées dans les plaques de culture cellulaire à 96 puits et ont été cultivées pendant 24 heures. Les cellules ont été cultivées pendant 24 heures, puis incubées avec différentes concentrations de MPP (un antagoniste spécifique de ERα, MPP dihydrochloride) pendant 1h, et ensuite, de la silibinine à 200µM a été ajoutée et cultivée pendant 24 heures. Les cellules ont été rincées deux fois avec PBS glacé et incubées avec 100µL d'une solution MTT à 0,5mg/mL à 37°C pendant 3 heures. |

Modèles animaux | Modèle de rats portant une tumeur sous-cutanée de cellules SKOV3 |
Méthode de préparation | Un groupe de rats nus BALB/c féminins âgés de 4 semaines a été anesthésié et ovariectomisé bilatéralement. Les cellules SKOV3 ont été récoltées et resuspendues dans un gel matriciel. Des cellules (5×106) ont été inoculées par voie sous-cutanée dans le flanc droit des rats ovariectomisés Les rats portant des xénogreffes d'environ 5mm de diamètre ont été aléatoirement divisés en trois groupes de cinq rats chacune. Chaque groupe a été traité avec véhicul, DPN (1mg/kg par jour) ou MPP dihydrochloride (1mg/kg par jour), par injection intra-péritonéale tous les jours. |
