ALV2はHelios(IKZF2)の効果的で選択的な分解剤である。ALV2は、CRBNに結合し、IC50値は0.57μMである[1-2]。Heliosは亜鉛フィンガー転写因子であり、炎症腫瘍微小環境で安定した制御性T細胞(Treg)の表現型を維持する。ALV2は、T細胞機能、腫瘍免疫微小環境の制御の研究に使われる[3-4]。
In vitro(体外)実験で、ALV2(1μM)でヒトナイーブCD4+ T細胞を5日間前処理し、その後は抗CD3/CD28抗体で2日間活性化した。この処理は、CD25とエフェクターサイトカイン(IL-21やIFNγなど)の産生を著しく上方制御し、ヒト滑膜組織マウスキメラモデルで、炎症組織を浸潤するこれらの細胞の能力を増強した[5]。サルモネラ(Salmonella)に1時間感染させたRAW 264.7マウスマクロファージで、ALV2(10μM;24時間の処理)は、腫瘍壊死因子α(TNF-α)の分泌を著しく減らした[6]。
In vivo(体内)実験で、ヒト化CrbnI391V遺伝子型のマウスモデルに、ALV2を腹腔内注射(100mg/kg;毎日二回;7日間)したところ、Ikaros(IKZF1)のレベルを影響せず、脾臓CD4+ FoxP3+制御T細胞(Tregs)でHelios(IKZF2)のタンパク質レベルを著しく低下させた[7]。
