GB1107はガレクチン-3(Gal-3;Kd=37nM)の経口活性の阻害剤である[1-2]。GB1107は抗ウイルスと抗癌特性を持ち、肺癌、胃癌、肝臓癌など様々な癌や椎間板変性に関連する研究に使える[3-4]。
In vitro(体外)実験で、FTC-133と8505C甲状腺癌細胞をGB1107(10–100μM)で24時間処理した。AKTのリン酸化を軽減させ、β-カテニンとMMP2の発現を下方制御することで、GB1107は細胞の接着を著しく阻害し、アノイキスを誘発し、同時に細胞の遊走と浸潤を用量依存的に減らした[5]。野生型マウス骨髄に由来するマクロファージ(WT BMDM)をGB1107(5μM)で24時間前処理し、その後はLLC腫瘍細胞条件付け培地(LLC CM)で刺激した。GB1107は、腫瘍細胞に対するマクロファージの貪食能力を増強させ、免疫抑制性M2様の表現型に対するそれらの分極を抑制した[6]。
In vivo(体内)実験で、骨髄系細胞特異的なTIM-3ノックインマウス(FSF-TIM3/LysM-Cre+/-)には、7週齢から10週齢までの間に、GB1107(5mg/kg)を隔日で腹腔内注射した。GB1107は、肺の炎症を著しく軽減させ、肺部炎症性サイトカインのIL-1βとTNF-αの転写レベルを下方制御させた[7]。CCl₄誘発性肝臓線維症モデルのC57BL/6Jマウスに、モデルの最後の4週間に、GB1107(10mg/kg)を毎日一回経口投与した。GB1107は肝臓線維症を著しく軽減し、その証拠に、ピクロシリウスレッドで染色された面積が減少し、ALTやASTなどの肝臓損傷の血漿マーカーが低下した[8]。
