LDS 751は、細胞膜透過性核酸染色剤であり、無核細胞と損傷された核細胞から無傷の核細胞を区別する為に使われ、フローサイトメトリーで混合された好中球、リンパ球と単球で異なる細胞タイプを区別する為にも使われている。LDS751の励起/発光光はそれぞれ543/712nmである(DNAと結合する場合)。

LDS 751は、細胞膜透過性核酸染色剤であり、無核細胞と損傷された核細胞から無傷の核細胞を区別する為に使われ、フローサイトメトリーで混合された好中球、リンパ球と単球で異なる細胞タイプを区別する為にも使われている。LDS751の励起/発光光はそれぞれ543/712nmである(DNAと結合する場合)。DsDNAと連結するLDS 751の最大励起波長は~543nmであり、488nmのアルゴンイオンレーザーで励起されることが可能である。その最大発光波長がより長い(~712 nm)為、マルチカラー分析には特に適宜である。DsDNAとの連結は、~20倍の蛍光増強を引き起こす。チアゾールオレンジ(主にRNA、GC30038と連結)とLDS751(主にDNAと連結)は赤血球、血小板、網状赤血球と有核細胞を区別する為に使われる。
このプロトコールはあくまでもガイドであり、お客様にはご自分のニーズに合わせてご調整願います。
1.LDS751染色溶液の調製
(1) LDS 751染色貯蔵液の調製:5.297mLのDMSOを使って、25mgのLDS 751を完全に溶解し、10mMの貯蔵液を獲得する。
注:LDS751貯蔵液は、小分けの後、繰り返した冷凍と解凍を避ける為に、暗闇で-20℃で貯蔵することを推奨する。
(2)LDS 751作業溶液の調製:事前加熱した無血清培養基または緩衝液(例えばHBSSまたはPBS)を使って、貯蔵液を薄め、10μMの濃度でLDS 751作業溶液を調製する。
注:実際の条件に応じてLDS751作業溶液の濃度を調整して、使用する前に調製してください。
2.細胞の染色
2.1懸濁細胞(6ウェルプレートを例に挙げて)
(1) 懸濁細胞を1000gで3-5分間遠心分離する。上清を廃棄し、PBSで5分間ずつ、計2回洗浄する。
(2) 1mLのLDS751作業溶液を入れて、暗闇で15-60分、室温で培養する。
(3) 培養の後、1000gで5分間遠心分離し、上清を除去し、PBSで5分間、2-3回洗浄する。
(4) 細胞を無血清の培地またはPBSで再懸濁し、蛍光酵素標識プレートまたはフローサイトメトリーで観察する。
2.2 接着細胞
(1) 接着細胞を無菌カバーガラスに培養する。
(2) 培養基からカバーガラスを除去し、余分な培養液を吸引しカバーガラスを湿った環境で置く。
(3) 100uLのLDS751作業溶液をカバーガラスの一角から入れて、染料が全ての細胞をカバーできるようにゆっくり振って、暗闇で室温で15-60分間培養する。
(4) 色素作業溶液を吸引し、培養基でカバーガラスを二三回洗浄し、蛍光酵素標識プレートまたはフローサイトメトリーで観察する。
3.蛍光ELISAプレートの検出:LDS751の励起/発光光はそれぞれ543/712nmである(DNAと結合する場合)
注:
(1)固定された細胞または組織サンプルを染色する為に、固定後、固定液を濯いで除去する;
(2)蛍光消光を遅らせる為に、抗蛍光消光シーリング剤を使うことを推奨する;
(3)LDS751は人間の体に刺激的である。安全と健康の為には、実験中は白衣と使い捨ての手袋を装着してください。
(4)最初の実験では、複数の染色濃度を試し、最適の作業濃度を決める。より高い濃度は細胞組織の非特定的な染色を起こす可能性がある。
