トリパリマブ（Toripalimab）は、組換えヒト化抗PD-1モノクローナル抗体である。トリパリマブはT細胞表面のPD-1受容体に結合し、PD-1とリガンドのPD-L1およびPD-L2との相互作用を阻害することで、PD-1シグナル伝達経路を介した免疫抑制を軽減する。トリパリマブは、悪性黒色腫、尿路上皮がん、鼻咽頭がん、食道扁平上皮がんなどのがんに関する研究に使用することができる。
In vitroでは、SEBで刺激されたヒト末梢血単核球（PBMC）をトリパリマブ（3.3～10 µg/mL）で3日間処理した場合、あるいは抗CD3/抗CD28で刺激されたヒトナイーブCD8+ T細胞を3日間処理した場合。トリパリマブは、Th1サイトカイン（IFN-γ、IL-2、TNF）および骨髄由来の炎症性サイトカイン（IL-1α、IL-1β）の分泌を著しく増強するとともに、T細胞の活性化も促進した。NSCLC患者から分離した腫瘍細胞のex vivo培養において、トリパリマブ（10 µg/mL）による処理と抗CD3/抗CD28刺激を6～24時間併用すると、IFN-γの産生および免疫細胞経路に関連する遺伝子の発現が有意に上昇した。ヒト末梢血単核細胞（PBMC）をトリパリマブ（0.01～10 µg/mL）で6～10日間処理したところ、トリパリマブは用量依存的にヒトT細胞の増殖を刺激し、IFN-γおよびTNF-αの分泌を増強した。
In vivoでは、MC38腫瘍を移植したC57/hPD-1ノックインマウスに対し、トリパリマブ（0.3～10mg/kg、腹腔内注射、週2回）を3週間投与した。トリパリマブは用量依存的に腫瘍の増殖を有意に抑制した。B16-vecまたはB16-IL33メラノーマ細胞を皮下接種したhPD-1ノックインマウスに対し、トリパリマブ（200µg/100µL、週2回、3週間）を投与した。トリパリマブとIL-33の併用により、腫瘍の増殖が有意に抑制され、腫瘍を有するマウスの生存期間が延長された。

調製方法 |
ヒト末梢血単核細胞（PBMC）を単離し、IL-4およびGM-CSFの存在下で6日間培養して未熟樹状細胞（imDC）を作製した。その後、imDCをTNF-αとともに3日間インキュベートし、成熟樹状細胞（mDC）を得た。同一ドナーのPBMCからT細胞を単離し、CFSEで標識した。mDC、T細胞、および破傷風トキソイド（10 ng/mL）を混合した。この混合液に、異なる濃度のトリパリマブ（0.01～10 μg/mL）、オプジーボ（陽性対照）、またはhIgG4（陰性対照）を添加した。 |
