Triptorelin ([DTrp6]-LH-RH)はゴナドトロピン放出ホルモン（GnRH）の高い選択性の作動剤である。TriptorelinはGnRH受容体と結合し、ゴナドトロピンの放出を刺激し、よって性ホルモンのレベルを調節する[2]。Triptorelinは前立腺癌、子宮内膜症や子宮筋腫など様々な性ホルモン関連の疾患の治療に使われる[3]。Triptorelinは、思春期の発育のコントロールと早発性思春期の阻害にも使われる[4]。
体外で、Triptorelin peptide (Triptorelin-AuNPs)と共役した金ナノ粒子を、20μg/mlの濃度で、MCF-7乳癌細胞を24時間事前処理し、その後、2、4、6と8Gyの線量で、6MeV X線で放射した。Triptorelinは、細胞の放射線に対する感受性を著しく増強し、同時に細胞の生存率を減らし、アポトーシス率を増やした[5]。Triptorelin (10nM–10μM)でLNCaP前立腺癌細胞株を9日間治療した。TriptorelinはLNCaP細胞に対し、用量依存的な阻害作用を示した。Triptorelin (10μM)はG0/G1期で細胞周期停止を起こし、S期で細胞の割合を減らした[6]。
体内で、35日齢のKMマウスに、Triptorelin (10、20、40μg；毎日一回)を7日間連続皮下注射した。Triptorelinはマウスで、子宮の成長と発育を著しく阻害し、子宮の重量を減らし、子宮内膜上皮肥厚（EET）と子宮壁肥厚（UWT）を減らした[7]。マウスにシクロホスファミド（75mg/kg）と/またはドキソルビシン（7.5mg/kg）を投与した後、Triptorelin (1mg/kg)を15日間連続腹腔内注射した。マウスの子宮卵巣組織に対するシクロホスファミドと/またはドキソルビシンの破壊性の影響を、Triptorelinは著しく減らした。Triptorelinは、原始卵胞、前胞状卵胞と顆粒膜細胞の数を著しく増やし、閉鎖性卵胞の数を減らした。Triptorelinは損傷を受けたグループで、卵巣と胞状卵胞内の卵母細胞の容積をも著しく改善した[8]。
