Description of Boc-LRR-AMC

Boc-LRR-AMC는 트립신을 위한 형광성 기질이고 20S 프로테아좀과 유사한다. Boc-LRR-AMC는 26S 프로테아좀이나 20S 프로테아제의 핵심에서 트립신 유사 활성을 검출하는데 사용할 수 있다. Boc-LRR-AMC의 검출 파장은 380 nm/460 nm이다[1].

Protocol of Boc-LRR-AMC

프로테아솜 검사 캡처를 위한CAPA (문헌에서, 필요에 따라 수정하십시오)[1][2].

1.항체 코팅 : 5µg/ml의 대상 항체를 준비하고 검은색 96웰 MaxiSorp 판을 코팅하고 2% BSA를 함유한 PBS로 1시간 동안 추가로 블로킹합니다.

3. 기질 배양 : 프로테아좀을 포획한 후 판을 먼저 20 mM트리스 버퍼(0.1% NP40을 함유하고 있고 pH 7.5)로 신중하게 세척하고20 mM Tris 버퍼(pH 7.5)로 세척하고 과잉물질을 제거합니다. 이후에 20 mM 트리스 버퍼(pH 7.5)로 희석된 프로테아솜 억제제를 웰에 추가하고 실온에서 10분 동안 회전하고 배양합니다. 마지막으로 형광 기질 Boc-LRR-AMC를 20 mM 트리스 버퍼로 희석하고 100 µM의 농도로 웰에 추가합니다.

4. 형광 검출 : 판을 플라스틱 커버로 밀봉하고 37°C에서 약 30분 동안 배양했습니다. 이 후에 Proteasome-GloMAX 또는 SpectraMax 190 마이크로플레이트 리더를 사용하고 흥분 파장 380 nm와 방출 파장 460 nm에서 형광 값을 측정했습니다.
