BODIPY 500/510 C1, C12 is a fluorescent fatty acid analog with maximum excitation/emission wavelengths at 500/510nm[1]. BODIPY dye is a small molecule dye with strong ultraviolet absorption ability. Its fluorescence peak is relatively sharp and its quantum yield is high. It is relatively insensitive to the polarity and pH value of the environment and is relatively stable under different physiological conditions[2]. Due to its asymmetric structure, BODIPY has a variety of derivative structural products[3]. BODIPY lipid droplet dyes can easily pass through the cell membrane and enter the cell interior. They can be specifically stained by localizing on neutral lipids in the cell, so the dye can be used to label living cells and fixed cells[4, 5].

1. Preparation of staining solution
(1) Prepare stock solution: Dissolve BODIPY 500/510 C1, C12 in DMSO to prepare a stock solution with a concentration of 1-10mM.
Note: Store unused stock solution at -20°C or -80°C away from light to avoid repeated freezing and thawing.
(2) Prepare working solution: Dilute the stock solution with a suitable buffer (such as serum-free medium or PBS) to prepare a working solution with a concentration of 1-10μM.
Note: Please adjust the working solution concentration according to actual conditions and prepare it before use.
2. Cell suspension staining
(1) Suspended cells: Centrifuge at 1000g for 5min at 4°C, discard the supernatant, and wash twice with PBS for 5min each time.
(2) Adherent cells: Wash twice with PBS, add trypsin to digest the cells, and centrifuge at 1000g for 3-5min after digestion.
(3) Resuspend approximately 106 cells in 1mL of BODIPY 500/510 C1, C12 working solution and incubate at 37°C in the dark for 5-30min. The optimal incubation time varies for different cells. Please explore according to the specific experimental requirements.
(4) After incubation, centrifuge at 1000g for 5min, remove the supernatant, and wash 2-3 times with PBS, each time for 5min.
(5) Resuspend the cells in pre-warmed serum-free cell culture medium or PBS and observe by fluorescence microscopy or flow cytometry.
3. Cell adhesion staining
(1) Culture adherent cells on sterile coverslips.
(2) Remove the coverslip from the culture medium, aspirate the excess culture medium, and place the coverslip in a humid environment.
(3) Add 100μL of dye working solution from one corner of the coverslip and gently shake to evenly cover all cells with the dye.
(4) Incubate at 37 °C in the dark for 5-30min. The optimal incubation time for different cells is different. Please explore it yourself according to the specific experimental needs.
(5) After the incubation, aspirate and discard the dye working solution, and wash the coverslip 2-3 times with pre-warmed culture medium.
Notes:
(1) Please adjust the concentration and incubation time of BODIPY 500/510 C1, C12 working solution according to the actual situation.
(2) It is recommended to perform a positive control in the experiment, such as incubating the control group cells with 30μM oleic acid for 8h before conducting subsequent experiments.
