Description of DBCO-NHCO-PEG4-amine

DBCO-NHCO-PEG4-amine is a PEG-based PROTAC linker can be used in the synthesis of PROTACs. DBCO-NHCO-PEG4-amine is a cleavable ADC linker used to conjugate MMAE and antibody (e.g., DBCO-VCpAB-MMAE and DBCO-TRX-MMAE with EC50s of 280 nM and 22 nM in SKBR3 cells, respectively)[1].

PROTACs contain two different ligands connected by a linker; one is a ligand for an E3 ubiquitin ligase and the other is for the target protein. PROTACs exploit the intracellular ubiquitin-proteasome system to selectively degrade target proteins.
ADCs are comprised of an antibody to which is attached an ADC cytotoxin through an ADC linker.
