Lyso-Tracker Red is a lysosome red fluorescent probe that can permeate cell membranes and can be used for lysosome-specific fluorescent staining of living cells.
Lyso-Tracker Red is a weakly basic fluorescent probe that is fluorescently labeled with DND-99 from Molecular Probes. Only the weak base can partially provide protons to maintain the pH at neutral and can be selectively retained In acidic lysosomes, specific fluorescent labeling of lysosomes can be realized. Both Neutral Red and Acridine Orange can also stain lysosomes, but the staining of Neutral Red and Acridine Orange lacks specificity. Lyso-Tracker Red is suitable for fluorescent staining of lysosomes in living cells, but not suitable for fluorescent staining of lysosomes in fixed cells. Refer to Figure 1 for the chemical structure of Lyso-Tracker Red molecule.

The excitation and emission spectra of Lyso-Tracker Red. Lyso-Tracker Red is an eosinophilic fluorescent probe for labeling and tracing acidic organelles in living cells. These probes have several important features, including highly selective targeting of acidic organelles and effective labeling of living cells at nanomolar concentrations. Lyso-Tracker Red must be in a very low concentration (usually about 50nM) to obtain excellent selectivity. Although the retention mechanism of these probes has not been clearly studied, it is probably related to the protonation and retention of acidic organelles. The endocytosis kinetics study of the Lyso-Tracker Red probe shows the uptake of dye into living cells. It only takes a few seconds. However, these lysosomal probes will cause the lysosome to be alkalized, and long-term incubation will induce an increase in the pH of the lysosome. Therefore, it is recommended to incubate the cells with the probe for not too long before imaging. The specific use concentration and incubation time of Lyso-Tracker Red probes need to be explored according to their own experimental conditions and specific cell types to achieve satisfactory staining results. Refer to Figure 3 for the staining effect of Lyso-Tracker Red in living cells.

Lyso-Tracker Red is suitable for fluorescent staining of lysosomes in living cells, but not suitable for fluorescent staining of lysosomes in fixed cells. If the cells stained with Lyso-Tracker Red need to be fixed, try 3% glutaraldehyde.
Dilute according to the ratio of 1:20000 to prepare 1000ml Lyso-Tracker Red working solution.
