Pyridoxal phosphate is the active form of vitamin B6 and can inhibit reverse transcriptases. Pyridoxal phosphate acts as a coenzyme that catalyzes amino acid metabolism reactions such as transamination, decarboxylation, and racemization by forming Schiff bases and stabilizing carbanion intermediates. Pyridoxal phosphate is used in research on pyridoxal phosphate-dependent enzyme-related disease prevention and tardive dyskinesia treatment[1-4].
In vitro, treating MMQ, GH3, and AtT-20 cells with 10-1000μM Pyridoxal phosphate for 2-4 days dose-dependently reduced cell number, arrested the cell cycle at G0-G1 phase, induced apoptosis, and inhibited GH and PRL secretion from GH3 cells and ACTH secretion from AtT-20 cells[5]. Treating bFGF-stimulated HUVEC cells with 50-250μM Pyridoxal phosphate for 72 hours dose-dependently inhibited cell proliferation and suppressed the activities of replicative DNA polymerase α and DNA topoisomerases I and II[6]. Treating HEK293 cells stably expressing P2X2 receptors with 7.3-13μM Pyridoxal phosphate for 5-10 minutes blocked ATP-evoked inward currents through P2X receptors[7].
In vivo, injecting 20mg/kg Pyridoxal phosphate intraperitoneally into offspring of prenatal Tebufenpyrad-exposed C57BL/6j mice from postnatal day 21 to 35 upregulated Pdxk and Gad1 expression in the brain and restored anxiety-like behaviors[8]. Injecting ddY mice intraperitoneally with a single dose of 5% Pyridoxal phosphate solution at 1% body weight volume induced tonic-clonic convulsions and death[9]. ICR mice injected intracerebrally with Aβ25-35 into the lateral ventricle were given 1mg/kg of Pyridoxal phosphate intraperitoneally for 14 consecutive days. Pyridoxal phosphate improved spatial and object cognitive function, reduced brain malondialdehyde and nitric oxide levels, downregulated the Bax/Bcl-2 ratio and cleaved caspase-3 protein expression, and decreased APP, PS1, and PS2 protein expression[10].

Preparation Method |
MMQ, GH3 and AtT-20 cells were maintained in RPMI-1640 or DMEM with serum, starved in phenol-free medium with 0.3% BSA for 6-12h, then treated with Pyridoxal phosphate at 10-1000μM for 2-4 days. After treatment, cell number, [3H]-thymidine incorporation, hormone (GH/PRL/ACTH) secretion, cell-cycle distribution, apoptosis (Annexin V-FITC), and Bcl-2 protein content were measured. |
