Iristectorin B est une isoflavone naturelle issue de Iris tectorum dotée d'effets anticancéreux[1]. Iristectorin B exerce une activité anti-inflammatoire par voie d'inhiber la production de NO induite par lipopolysaccharide (LPS)[2]. Iristectorin B est largement utilisé en tant que référence interne pour développer des méthodes d'extraction assistées par ultrasons dans le liquide ionique pour l'extraction et la séparation de composés connexes[3].
Le traitement in vitro avec Iristectorin B (25µM) pendant 24h a significativement amélioré la survie de cellules PC12 après un traitement de privation de glucose, d'oxygène/réoxygénation (OGD/R), réduit l'apoptose cellulaire et l'expression de TFR1, HMOX1, et SLC3A2[4].

Méthode de préparation | Les cellules PC12 ont été cultivées dans un milieu DMEM à haut glucose supplémenté avec 10% de sérum de veau foetal (FBS) et 1% de pénicilline/streptomycine à 37°C en présence de 5% CO2. 100μl de solution EBSS sans sucre contenant 10mM de Na2S2O4 ont été ajoutés et incubés pendant 2h. Les cellules ont été remplacées par un milieu normal pour la réoxygénation et la culture a été poursuivie pendant 24h, constituant un modèle d'agression OGD/R induit pour simuler une lésion cérébrale ischémique. OGD/R a été supplémenté avec un milieu de culture normal contenant de Iristectorin B à 6.25µM, 12.5µM, et 25µM pendant 24h. La viabilité cellulaire a été analysée. | ||
