MEGA-8 est un tensioactif non ionique de type glucoside, obtenu par la condensation d'un groupe octanoyle (chaîne acyle linéaire en C8) avec la N-méthylglucamine[1]. MEGA-8 se caractérise par sa douceur et sa nature non dénaturante. Sa concentration micellaire critique (CMC) est pratiquement non affectée par la température entre 5 et 40 °C, elle est de 70 mM en l'absence de sels, et comprise entre 5 et 64 mM dans des conditions de forte et de faible salinité[2][3]. MEGA-8 est généralement utilisé dans la préparation de biomatériaux ou de composé organiques, et on le rencontre couramment dans la recherche en sciences de la vie, comme pour l'extraction de protéines et la solubilisation des protéines membranaires[4][5]. Lorsqu'il est associé à CHAPSn MEGA-8 réduit sa CMC de manière synergique et améliore son activité de surface, permettant ainsi un solubilisation efficace des protéines membranaires dans des conditions douces[6]. MEGA-8 déplace délicatement PrBP/δ et inhibe l'activité basale de PDE6, tout en préservant l'intégrité et l'homogénéité du complexe holoenzymatique. Il constitue donc un environnement détergent idéal pour la purification et les études structurales de la phosphodiestérase 6 rétinienne[7].
