Cryptotanshinoneは丹参(Salvia miltiorrhiza Bunge)の根から抽出された天然化合物であり、シグナル伝達と転写活性化因子3(STAT3)を阻害することで、抗腫瘍活性を示し、IC50値は4.6μMである[1, 2]。Cryptotanshinoneは、抗腫瘍、抗炎症、抗線維症と抗血管新生効果を含む複数の生物活性を持っている[3, 4]。
体外では、Cryptotanshinone (20μM)でA549細胞を24時間処理した結果、G0/G1期にある細胞の割合を減らし、G2/M期における細胞の割合を増やし、G2/M期の停止を誘発し、アポトーシスの遺伝子の発見と共にアポトーシスを促進した[5]。もう一つの研究では、Cryptotanshinone (3μM)でH9C2細胞を1時間事前処理した結果、長期間低酸素誘発の細胞損傷を著しく阻止し、細胞ATPレベルを復元し、ミトコンドリアスーパーオキシドのレベルを減らし、細胞内NOとカルシウムレベルを減らした[6]。
体内では、大動脈バンディング(AB)手術に誘発された心臓リモデリングのマウスモデルに、Cryptotanshinone (50mg/kg/日)を4週間経口投与した結果、圧力オーバーロード誘発の心臓障害を改善し、心臓肥大を軽減し、心臓線維症を減らした[7]。更に、Cryptotanshinone (40mg/kg/日)をCCl4誘発の肝臓線維症マウスモデルに経口投与した結果、肝臓線維症を改善し、肝星細胞の活性化を阻害し、コラーゲンの製造を抑制した[8]。

準備方法 | 1x105/mlの密度で、A549細胞を6ウェルのプレートに播種し、最終濃度が20μMのCryptotanshinoneで24時間処理した。細胞をトリプシンで処理して、遠心分離(4℃、800g/分、10分間)し、PBSで洗浄し、4℃で冷たい70%のエタノール/30%のPBSで一晩中固定した。細胞をRNase Aで消化し、その後、37℃で1%のヨウ化プロピジウムで30分間染色した。染色の4時間内で、DNAプロフィールをフローサイトメトリーで測定した。細胞周期とアポトーシス関連の遺伝子の発見を、Cell Cycle PCR Arrayでテストした。 |

準備方法 | 大動脈バンディング(AB)手術の後、マウスを4つのグループにランダムに分けた:生理食塩水(VEH)、Cryptotanshinone (CTS)グループ、ABグループ、AB+CTSグループ(模擬実験グループにn=10、ABグループにn=10)。AB手術の3日後に、マウスにCryptotanshinoneを4週間投与(50mg/kg、胃潅流)した。処理の終わりに、生存したマウスを、心臓機能の評価に使った。そしてマウスにペントバルビタールナトリウム(200mg/kg、腹腔内注射)を過剰投与することで、安楽死させた。他の実験のために、心臓を採取した。 |
