Description of Cauloside A

Cauloside A는 Dipsacus asper 및 Caulophyllum thalictroides 등의 식물에서 분리된 트리터펜오이드 사포닌으로 항염증 및 항진균 활성을 보유하고 있다. Cauloside A는 미토콘드리아 막 완전성을 파괴하고 미토콘드리아 기능을 손상시켜서[3] 식물 진균성 질병 방제 연구 및 항염증와 항종양 약물 개발에 일반적으로 사용된다[4,5].

체외 실험에서 전식물 검정법을 사용해서 Cauloside A (500μg/mL)을 벼와 토마토 잎에 분무한 후 24시간이 지난 후 해당 병원체(도열병(RCB), 토마토 잿빛곰팡이병(TGM), 토마토 역병(TLB))를 접종하였다. RCB, TGM 및 TLB에 대한 방제 효과는 각각 95%, 83%, 97%에 달하였다[6]. Cauloside A (1, 5, 10, 50μg/mL)과 지방다당류 (LPS, 100ng/mL)를 BV2 미세아교세포에 24시간 동안 공동 처리하면 LPS 유도 NO 생성을 용량 의존적으로 억제하였고 50μg/mL 농도에서 가장 현저한 억제 효과가 관찰되었다[7].

체내 실험에서 ICR 쥐를 Cauloside A (200mg/kg)을 함유한 C. thalictroides 조제물로 경구 전처리한 후 30분간간 LPS (1mg/kg)을 복강 내 주사해서 추가 24시간 처리하였고 부신 조직에서 LPS 유도 COX-2, iNOS, TNF-α, IL-1β 및 IL-6의 mRNA 발현을 현저하게 억제하였다[7].

제조 방법

BV2 세포를 24웰 판에 웰당 1×10⁵ 개의 세포 밀도로 배양하였고 LPS (100ng/mL) 존재 하에 다양한 농도 (1, 5, 10, 50μg/mL)의 Cauloside A로 24시간 처리하였습니다. 배양 후 50μL의 배양 상청액을 수집해서 동일 부피의 그리스 시약과 혼합하였습니다. 효소 표지로 540nm에서 흡광도를 측정하고 아질산염 축적량을 평가해서 NO 생성을 평가하였습니다.

반응 조건

1, 5, 10, 50μg/mL; 24 시간 동안

응용 분야

Cauloside A 처리는 LPS 유도 NO 생성을 용량 의존적으로 억제하였고 50μg/mL 농도에서 가장 현저한 억제 효과가 관찰되었습니다.

ICR 쥐를 Cauloside A를 함유한 C. thalictroides 조제물 (200mg/kg)으로 경구 전처리한 후 30분간 LPS (1mg/kg)을 복강 내 주사해서 추가 24시간 처리하였고 이후 PCR을 통해 부신 조직에서 COX-2, iNOS, TNF-α, IL-1β 및 IL-6의 mRNA 발현 수준을 분석하였습니다.
