LX-4211은 경구 투여용 강력한 이중 SGLT2/1 억제제로 SGLT1과 SGLT2에 대한 IC50 값은 각각 36nM과 1.8nM이다. LX-4211은 SGLT2를 억제해서 요당 배설을 증가시키고 SGLT1을 억제해서 위장관의 포도당 흡수를 줄여서 혈당 수준을 효과적으로 낮춘다[3][4]. LX-4211은 당뇨병 관련 연구에서 일반적으로 사용된다[5][6].
체외 실험에서 돼지 관상동맥 배양 내피세포(ECs)를 LX-4211(10nM)로 30분간 처리한 후 정상 포도당 또는 고 포도당(HG: 25mmol/L)에서 24시간 동안 추가 배양하면 고 포도당에 의해 자극된 포도당 흡수가 감소하고 ECs 노화표지자와 산화적 스트레스가 줄어들고 eNOS 발현과 NO 생성이 상향 조절되고 VCAM-1, 조직 인자 및 국소 르네신-안지오텐신 시스템의 발현이 감소한다[7].
체내 실험에서 T1D를 유도한 쥐를 대상으로 LX-4211(경구 투여; 2/30mg/kg/일)을 22일 동안 투여하면 혈당 수준과 A1c 수준이 현저하게 감소하고 저혈당 측정 비율은 증가하지 않는다[8]. 쥐에게 LX-4211(60mg/kg)을 경구 투여하면 SGLT1을 억제해서 장내 포도당 흡수를 줄이고 글루카곤 유사펩타이드-1(GLP-1)과 펩타이드 YY(PYY)의 방출은 증가시키고 포도당에 의존하는 인슐린 폴리펩타이드(GIP)의 방출과 포도당 함유 식사 후 6시간 동안 혈당 수준의 변화는 줄인다[4].
