Description of Triptolide

Triptolide es un triepóxido de diterpeno aislado por primera vez de la planta medicinal Tripterygium wilfordii, que exihibe efectos inmunosupresores, antiinflamatorios, antiproliferativos y antitumorales. Actúa como un inhibidor de la activación de NF-κB, donde NF-κB es una familia de complejos de proteínas del factor de transcripción[1]. Triptolide posee una actividad anticancerígena de amplio espectro, inhibiendo el desarrollo del cáncer de colon, el cáncer de mama, el carcinoma de cálulas renales y el cáncer de cuello uterino[2].

In vitro, triptolide (50 nM, 72 h) inhibe significativamente la proliferación de células de cáncer de ovario SKOV3 y A2780. Triptolide (15 nM, 48h) inhibe significativamente la migración celular y bloquea la capacidad invasiva de las células[2]. Triptolide (50-200 nmol/L) incubado con células PANC-1 y MiaPaCa-2 reduce significativamente la viabilidad celular sin afectar la viabilidad de las células ductales pancreáticas normales [3]. El valor IC50 de triptolide en la viabilidad celular de RSF es de 74.3 nM, y su valor de IC50 en la proliferación de células de RSF es de 20.4 nM[4]. Triptolide (0-150 nM) inhibe significativamente la viabilidad de las células HK1, FaDu, y C666-1 de una manera dependiente de la dosis y el tiempo[5].

In vivo, el tratamiento con triptolide (1 mg/kg/d, p.o.) durante tres semanas en ratones desnudos redujo el número de nódulos metastásicos en aproximadamente un 80%[2]. Triptolide (0.2 mg/kg/d) administrado a ratones con tumores pancreáticos trasplantados durante 60 días redujo el crecimiento del cáncer de páncreas y disminuyó significativamente la propagación local del tumor[3].

Las células SKOV3 y A2780 se chaparon en placas de 96 pozos (5 × 103/pocillo). 24h después, se añadió triptolide a una concentración final de 0, 1.5, 5, 15, 50, o 150 nM durante 72 h.

Triptolide (50nM, 72 h) inhibió notablemente la proliferación de células de cáncer de ovario. Triptolide (15nM) iinhibió significativamente la migración celular 48 horas después de la herida. Triptolide (15nM) bloqueó notablemente la capacidad invasiva de las células SKOV3 y A2780.

Método de preparación
