Description of Boc-Lys(Ac)-AMC

Boc-Lys(Ac)-AMC is a substrate of histone deacetylases (HDACs). HDACs catalyze the deacetylation of Boc-Lys(Ac)-AMC and through addition of a trypsin-containing developer, the deacetylated product can convert to a quantifiable fluorophore (Ex/Em=355nm/460nm) [1].

Protocol of Boc-Lys(Ac)-AMC

Ⅰ. Tissue culture-based HDAC assays [1]

1. Seed 6×104 cells in 100μl medium in 96-well tissue culture plates and incubate it overnight.

2. Add 25μM HDAC substrate Boc-Lys(Ac)-AMC into the overnight culture and incubate it at 37℃ with 5% CO2 atmosphere for 2~3h.

3. Add HDAC developer solution containing cell lysis buffer and trypsin reagents to terminate the deacetylation reaction.

4. After 15min incubation at room temperature, measure the fluorescent signal of AMC by microplate reader at Ex/Em=355nm/460nm.
