Description of Phleomycin

Phleomycinは糖ペプチドの抗生物質とDNA鎖切断(DNA-SB)の誘導剤であり、遺伝子組換え細胞の選択に使われることが可能である[1]。Phleomycinはストレプトマイセス属に発見された分子であり、ブレオマイシンに似ていて、DNAに結合、挿入でき、二重螺旋構造の完全性を阻害する[2]。Phleomycinは耐性連鎖球菌株に対し、変異原性作用を持つ[3]

試験管内、Phleomycin (100-400μg/mL)でHeLa細胞を0-24時間処理した結果、時間と用量依存的に細胞の増殖を阻害し、細胞毒性を誘発し、サイトクロムcの最初の放出、Bcl-XL、ENDOG、DAXXとMDM2の下方制御、そしてCASPとBIDの上方制御を誘発した[4]

Phleomycin (0.2μg/mL)でナンノクロロプシス・オーセアニカ細胞を48時間処理した結果、藻類細胞のDNAでランダムな二本鎖切断を起こし、活性ハイグロマイシンB耐性遺伝子(aph7)の変換効率を増やした[5]。様々な変異体からのマウス胎児線維芽細胞(MEF)をPhleomycin (0-200ng/mL)で48時間処理した結果、MEFを欠けるXRCC4、DNA-Ligase IVまたはXlfの生存率を著しく減らしたが、PAXXからのMEFと野生型のMEFに対してはこれと言った効果はなかった[6]

準備方法

細胞(1ウェルあたり6×104)を24ウェルプレートに1日間付着させ、Phleomycin を添加し、0, 6 , 12, 18, 24時間で、100, 200, 300, 400μg/mLのPhleomycinで血球計数器で測定した。HeLa細胞に対するPhleomycinの細胞毒性作用を、生細胞の数とMTTアッセイで測定した。

アプリケーション

Phleomycinは用量と時間依存的にHeLa細胞の増殖を阻害し、細胞毒性を誘発する。
