4-ethylphenyl sulfate is an orally active and blood-brain barrier-penetrating intestinal microbial metabolite [1]. N-4-Ethylphenyl sulfate acts as a protein-bound uremic toxin that causes endothelial dysfunction and leads to chronic kidney disease [2]. 4-Ethylphenyl sulfate is widely used as a diagnostic biomarker for intestinal flora imbalance and autism, and is used as a reference substance to develop liquid chromatography/electrospray ionization methods for the identification of related compounds[3].
In vitro, 4-ethylphenyl sulfate treatment (10mM) for 72 hours significantly reduced the viability of HCT-116 cells, accompanied by cell volume reduction, formation of intracellular vesicles, and damage to cell membrane integrity[4].

Preparation Method | HCT-116 cells were cultured at 37°C and 5% CO2 in McCoy 5A medium supplemented with 10% fetal bovine serum (FBS), 1% penicillin-streptomycin. Cells were plated at a density of 2.5×104 cells/ml in a 96-well plate with growth medium for 24h, and then were incubated with the different concentrations of 4-ethylphenyl sulfate (0, 1, 2.5, 5, and 10mM) for 24, 48, and 72h, respectively, the cell viability was analyzed. |
