Coelenterazine 400a, ein Derivat des natürlichen Coelenterazine[1] ,dient als Substrat für Renilla-Luciferase (Rluc)[2], mit einer maximalen Emissionswellenlänge von 400nm. In Gegenwart von Rluc reagiert Coelenterazine 400a mit molekularem Sauerstoff, bildet hochenergetische Zwischenprodukte und emittiert Licht[3][4]. Aufgrund seiner minimalen Störung von fluorescenten Akzeptoren ist es eine ideale Wahl für Biolumineszenzresonanzenergietransfer (BRET)-Assazs, die häufig zur Bewertung von Protein-Protein-Wechselwirkungen[5][6].

Experimentelles Protokoll für Coelenterazine 400a in BRET-Assay
1. Herstellung der Stammlösung: Lösen Sie Coelenterazine 400a in 100% Ethanol oder Methanol, um eine Stammlösung mit einer Konzentration von 1mM herzustellen. Aliquotieren Sie die Stammlösung und lagern Sie sie bei -20°C oder -80°C geschützt vor Licht.
2. Herstellung der Proben: Kultivieren Sie die Zellen in einem geeigenten Medium bis zu einer Konfluenz von 70-80%. Verwenden Sie die geeigneten Transfektionsreagenzien, um Zellen gemeinsam mit Renilla-Luciferase- und fluorescenten Acceptor-Plasmiden zu transfektieren. Kultivieren Sie weitere 24-48 Stunden nach der Transfektion , um eine ausreichende Proteinausdruckung zu gewährleisten.
1. Herstellung der Substratlösung: Vor der Verwendung verdünnen Sie die Coelenterazine 400a-Stammlösung mit Zellkulturmedium auf eine Konzentration von 100μM .
2. BRET-Assay: Waschen Sie die transfektierte Zellen und resuspendieren Sie in Phosphatpuffer-Lösung(PBS). 90µL der Zell Suspension in jede Vertiefung einer 96-Loch- weißen Mikroplatte geben. 10µL Substratlösung in jede Vertiefung geben, um eine endgültige Konzentration von 10µM zu erreichen.
3. Fluoreszenzmessung: Nach Zugabe von Coelenterazine 400a messen Sie die Luminanzintensität mit einem Multimode-Mikroplattenleser.
