RGFP966 ist ein N-(o-aminophenyl)-Carboxamid-HDAC-Inhibitor mit einem IC50-Wert von 0.08 μM für HDAC3. Es handelt sich um eine Klasse von kompetitiven, fest bindenden Inhibitoren mit langsamer Aktivierung oder Deaktivierung, die auf HDAC3 abzielen. RGFP966 unterdrückt nachweislich Entzündungsreaktionen bei verschiedenen entzündlichen Erkrankungen[1-3].
RGFP966(1 μM ;20 Stunden) schwächt die Expression entzündlicher Gene in LPS/IFNγ-stimulierten RAW 264.7-Makrophagen ab[3]. RGFP966(10-25 μM;48 Stunden) hemmte sowohl die Proliferation als auch die Migration von HCC-Zellen[4].
RGFP966(10 mg/kg; 6 Stunden vor SBI) hemmte die Hochregulation von HDAC3 und rettete die Nervenzellen im Bereich des geschädigten Bereichs bei Ratten mit chirurgischer Hirnverletzung(SBI)[5]. RGFP966(10 mg/kg/Tag; i.p; 5 Tage) konnte das LPS-induzierte depressive Verhalten bei Mäusen verbessern. Die Behandlung mit RGFP966 reduzierte die Expressionsniveaus von toll-like receptor 4 (TLR4), nucleotide-binding oligomerization domain-like receptor pyrin domain-containing-3 (NLRP3), caspase-1 und interleukin-1β (IL-1β)[6]. RGFP966(10 mg/kg oder 20 mg/kg) reduzierte die HDAC3-Expression und -Aktivität. Dann wurden die Rekrutierung von Eosinophilen und Mastzellen, die Proliferation von Becherzellen und die Konzentration inflammatorischer Zytokine verringert. Das führte zu einer Linderung allergischer und entzündlicher Reaktionen bei Mäusen mit allergischer Rhinitis[7].
