Z-DEVD-FMK ist ein specifischer irreversibler Cysteine-Asparagin-Protease 3 (caspase-3) inhibitor mit einer IC50 von18μM[1].Z-DEVD-FMK wird häufig zur Untersuchung der Apoptose und Neuroprotektion verwendet[2].
In vitro sendet das Behandlung von Kardiomioyztchen mit Z-DEVD-FMK (50μM) für 24 Stunden die zeramidinducede Apoptose signifikant und erhöhte die Zellüberlebensrate auf 81% [3]. Z-DEVD-FMK (15μM) kann die Aktivierung von Caspase-3 effektiv reduzieren, wenn zum Treatment von renalen proximalen Tubulizellen verwendet wird, jedoch ist die Wirkung bei der Senkung der zellulären Apoptose nicht so gut wie bei Pan-Capase-Inhibitoren [4].
In vivo verringerte das Treatment von Mäusen mit traumatischen Hirnverletzungen(TBI) mittels intrazerebralen Ventrikelinjrktions von Z-DEVD-FMK (160 ng) das Geschwürvolumen nach Verletztungen des Zentralnervensystems und verbesserte motorische und kognitive Funktionen [2]. Bei diabetischen Mäusen verbesserte eine intraperitoneale Injektion von Z-DEVD-FMK (20 mg/kg) über einen Zentraum von 8 Wochen die Proteinurie, die Nierenfunktion sowie die Tubulointerstitelle Fibrose und verlangsamte den Abfall der Serumalbuminkonzenration [5] . Z-DEVD-FMK (0.2 mg/kg) wird in den Augapfel injiziert, um eine Sehnerverschädung bei Kaninchen zu behandeln, wodurch die Apoptose von Netzhautganglienzellen signifikant reduziert und die Wiederherstellung der Sehnervfunktion gefördert wird [6].
