Description of FT895

FT895 est un inhibiteur sélectif et efficace de l'histone déacétylase 11 (HDAC11), sa valeur d'IC50 est 3nM[1].

HDAC11 est un nouveau membrane unique des histones déacétylase, ce qui est fortement exprimé dans les tissus spécifiques et certaines lignées de cellules cancéreuses[2]. HDAC11 possède à la fois les activités de déacétylation des histones et non-histones[3] et peut participer aux activités cellulaires comme le remodelage de la chromatine et les voies métaboliques de signalisation par réguler l'état d'acétylation de la cellule[3]. En tant qu'inhibiteur sélectif de HDAC11, FT895 a démontré les excellentes propriétés cellulaires et pharmacocinétiques[4], ce qui fait un outil efficace pour rechercher les mécanismes biologiques de HDAC11. D'ailleur, FT895 montre les applications prometteuses dans le traitement du cancer et des maladies inflammatoires.

Le traitement in vitro de FT895 (10μM) pendant 24 heures a entraîné une réduction significative de la capacité de migration des cellules A549 et H1650[5]. Le traitement de FT895 (10μM) pendant 1 heure a entraîné une amélioration significative de l'efficacité bactéricide dans les cellules BMDM (Macrophges dérivés de moelle osseuse)[6].

L'injection in vivo intra-péritonéale de FT895 (10mg/kg/jour) pendant 5 jours dans les rats C57BL/6J a promu l'expression du transporteur de la membrane mitochondriale interne UCP1 (Protéine de découplage 1) dans le tissu adipeux via l'inhibition de HDAC11, a entraîné ainsi finalement une augmentation de la température corporelle centrale des rats[7]. L'injection intra-péritonéale de FT895 (50mg/kg/jour) pendant 7 jours dans les rats C57BL/6, ce qui a augmenté la régulation de l'expression de la cytokine anti-inflammatoire IL-10 (Interleukin-10), ainsi donc, a promu les réponses anti-inflammatoires et a réduit la réplication du virus entérique EV71 dans les rats[8].

Méthode de préparation

Les cellules A549 et H1650 doivent être cultivées dans les plaques à 6-puits jusqu'à les fluorescences atteignent 90-95%. Ensuite, une pointe de pipette stérile de 2µl est utilisée pour céer une égratiguure dans chaque puits. Rincer les cellules avec PBS, puis, les incuber à 37°C pendant 24 heures dans un milieu supplémenté avec 10μM FT895. Finalement, les images du processus de cicatrisation de la blessure seront capturées à l'aide d'un système d'imagerie approprié pour l'observation et l'analyse.

Rats C57BL/6J

Méthode de préparation

Le nombre approprié de rats mâles C57BL/6J âgés de 6 semaines doit être préparé et placé dans les cages propres à ventilation individuelle. Ensuite, les rats seront infectés avec 1 × 106 PFU Gluc-EV71 dans 100 µl PBS pendant 7 jours. Simultanément, FT895 (50mg/kg/jour) sera également injecté par voie intra-péritonéale pendant 7 jours. Enfin, le nombre de copies du virus peut être déterminé par qPCR.
