FT895はヒストン脱アセチル化酵素11 (HDAC11)の効果的で選択的な阻害剤で、IC50値は3nMである[1]。HDAC11はヒストン脱アセチル化酵素ファミリーの新型でユニークなメンバーであり、特定の組織と一定の癌細胞株ではよく発見される[2]。HDAC11はヒストンと非ヒストン[3]脱アセチル化の活性の両方を持ち、細胞のアセチル化状態を調節することで、クロマチン更新や代謝シグナル経路などの細胞活動に関与できる[3]。HDAC11の選択的な阻害剤として、FT895は優れた細胞活性と薬物動態特性を示してきて[4]、FT895をHDAC11の生物学的なメカニズムの効果的な研究ツールにさせる。その他、癌と炎症の治療においても、FT895を応用することができる。
試験管内実験では、FT895 (10μM)で24時間処理した結果、A549とH1650細胞における移動能力が著しく減少した[5]。FT895 (10μM)で1時間処理した結果、BMDM(骨髄由来マクロファージ)細胞における殺菌効果が著しく増加した[6]。
生体内実験では、C57BL/6JマウスにFT895 (10mg/kg/日)を五日間腹腔内注射した結果、HDAC11の阻害により、脂肪組織におけるミトコンドリア内膜輸送体UCP1(脱共役タンパク質1)の発見を促進し、最終的にはマウスにおける体幹温度を上昇させた[7]。C57BL/6マウスにFT895 (50mg/kg/日)を七日間腹腔内注射した結果、HDAC11活性を阻害し、抗炎症サイトカインIL-10(インターロイキン-10)の発見を上方制御し、よってマウスにおいて抗炎症反応を促進し、腸管ウイルスEV71の複製を減少させた[8]。

準備方法 | A549とH1650細胞は90-95%の合流になるまで、6ウェルのプレートで培養する必要がある。その後、殺菌2μLピペットチップを使って全てのウェルに傷跡をつける。PBSで細胞を洗浄し、その後、10μMのFT895で補給された培地で37℃で24時間細胞を培養する。最後に、観察と分析の為に、傷の治癒過程の画像を適切なイメージングシステムを使って撮る。 |

準備方法 | 適切な数の6週齢のC57BL/6雄マウスは、綺麗で、個別換気ケージに置くべきである。その後マウスを七日間、100μLのPBSで1 × 106のPFU Gluc-EV71に感染させる。それと同時に、FT895(50mg/kg/日)を7日間腹腔内注射する。最後に、ウイルスのコピー数はqPCRで測定する。 |
