FT895 ist ein wirksamer selektiver Inhibitor der Histon-Deacetylase 11(HDAC11) mit einem IC50-Wert von 3 nM[1]. HDAC11 ist ein neuartiges und einzigartiges Mitglied der Histon-Deacetylase-Familie, das in bestimmten Geweben und bestimmten Krebszelllinien stark exprimiert wird[2]. HDAC11 hat sowohl Histon- als auch Nicht-Histon[3]-Deacetylase-Aktivitäten und kann an zellulären Aktivitäten wie Chromatin-Remodellierung und metabolischen Signalwegen beteiligt sein, dadurch dass es den Acetylierungszustand der Zelle reguliert[3]. Als selektiver Inhibitor von HDAC11 hat FT895 hervorragende zelluläre Aktivität und pharmakokinetische Eigenschaften gezeigt[4], was FT895 zu einem effektiven Instrument zur Untersuchung der biologischen Mechanismen von HDAC11 macht. Außerdem bietet FT895 vielversprechende Anwendungsmöglichkeiten in der Behandlung von Krebs und entzündlichen Erkrankungen.
In vitro führte die Behandlung mit FT895 (10μM) über 24 Stunden zu einer signifikanten Reduktion der Migrationsfähigkeit in A549- und H1650-Zellen[5]. Eine einstündige Behandlung mit FT895 (10μM) führte zu einer signifikanten Verbesserung der bakteriziden Wirksamkeit in BMDM-Zellen(Bone Marrow Derived Macrophage)[6].
In vivo förderte die intraperitoneale Injektion von FT895 (10mg/kg/Tag) über 5 Tage in C57BL/6J-Mäuse die Expression des mitochondrialen inneren Membrantransporters UCP1(Uncoupling Protein 1) im Fettgewebe durch Hemmung von HDAC11, was letztlich zu einem Anstieg der Körperkerntemperatur der Mäuse führte[7]. Die intraperitoneale Injektion von FT895(50mg/kg/Tag) über 7 Tage bei C57BL/6-Mäusen führte zur Hemmung der HDAC11-Aktivität, wodurch die Expression des entzündungshemmenden Zytokins IL-10 (Interleukin-10) hochreguliert wurde, wodurch entzündungshemmende Reaktionen gefördert und die Replikation des Enterovirus EV71 in den Mäusen verringert wurde[8].

Zelllinien | A549, H1650 |
Vorbereitungsmethode | A549- und H1650-Zellen müssen in 6-Well-Platten bis zu einer Konfluenz von 90-95% kultiviert werden. Dann wird mit einer sterilen 2 µl Pipettenspitze in jedes Loch ein Kratzer geritzt. Die Zellen werden mit PBS gewaschen und dann 24 Stunden bei 37°C in einem mit 10μM FT895 angereicherten Medium inkubiert. Abschließend werden Bilder des Wundheilungsprozesses mit einem geeigneten Bildgebungssystem zur Beobachtung und Analyse aufgenommen. |

Vorbereitungsmethode | Eine geeignete Zahl 6 Wochen alter männlicher C57BL/6-Mäuse wird vorbereitet und in saubere, einzeln belüftete Käfige gesetzt. Dann werden die Mäuse 7 Tage lang mit 1 × 106 PFU Gluc-EV71 in 100 µl PBS infiziert. Gleichzeitig wird FT895 (50mg/kg/Tag) auch 7 Tage lang intraperitoneal injiziert. Abschließend kann die Viruskopienzahl mittels qPCR bestimmt werden. |
