Description of 2,4-Dinitrobenzenesulfonic acid hydrate

2,4-Dinitrobenzenesulfonic acid hydrate is a derivative of benzenesulfonic acid, which can cause various inflammations in the body[1-2], and is commonly used as an inducer to establish animal models, such as colitis models[3]. 2,4-Dinitrobenzenesulfonic acid can be used as a catalyst for an amination reaction involving diols, and can also be used to synthesize the corresponding α-hydroxyketone[4].

In vivo, to establish a severe colitis mouse model, mice are anesthetized, and a 10-cm-long PE-90 catheter is inserted 3.5cm into the colon. A solution of 2,4-Dinitrobenzenesulfonic acid at a dose of 200mg/kg is injected through the catheter. On day 12, colitis is reactivated by a second injection of a 100mg/kg 2,4-Dinitrobenzenesulfonic acid solution. In the mild enteritis model, the catheter is placed using the same procedure, and the enteritis is reactivated by a second injection of 100mg/kg of 2,4-Dinitrobenzenesulfonic acid solution on day 21 after the first injection of 2,4-Dinitrobenzenesulfonic acid[5]. In order to establish a model of chronic enteritis, after anesthetizing the mice, a 10cm long PE-90 catheter was inserted into the colon of the mice at 3.5cm, and 2,4-Dinitrobenzenesulfonic acid solution was injected through the catheter at weekly injection doses of 2mg, 2mg, 4mg, 4mg, 6mg, 6mg, and 6mg for six consecutive weeks[6]. To establish a model of acute visceral pain, after a brief 2% isoflurane anesthesia, a PE-60 catheter was inserted into the rectum 8cm away from the anus, and 30mg of 2,4-Dinitrobenzenesulfonic acid dissolved in 0.25ml of 50% ethanol was injected into the body through the catheter for 14 to 21 days[7]. To establish a prostatitis model, mice are anesthetized, and a PE10 catheter sterilized with ethylene oxide and lubricated is inserted 1.5cm into the urethra. A volume of 40μl of a 10mg/ml 2,4-Dinitrobenzenesulfonic acid solution is injected, with symptoms being most pronounced on the second day after induction[8].

Protocol of 2,4-Dinitrobenzenesulfonic acid hydrate

Severe colitis model
1.Animal anesthesia:
Mice were anesthetized with 2% isoflurane.
2.Model establishment: A 10-cm-long PE-90 catheter was inserted 3.5cm into the colon. A solution of 200mg/kg 2,4 - dinitrobenzenesulfonic acid (dissolved in 30% ethanol) was injected through the catheter. The control group of mice received only 30% ethanol treatment. For the first 3 days after DNBS injection, the mice drank water containing 6% sucrose to prevent dehydration.
3.Reactivation of colitis: After a 4-day recovery period following the initial 2,4 - dinitrobenzenesulfonic acid injection, the mice were divided into several groups and were given specific bacterial strains or their culture supernatants via gavage for 7 days. On day 12 after the first DNBS injection, colitis was reactivated by a second injection of 100mg/kg DNBS solution.
