Picrotoxin(Cocculin) ist ein Pflanzen-Alkaloid, das als nicht-kompetitiver Antagonist von GABAA-Rezeptoren wirkt und deren Chloridkanal blockieren kann[1]. Picrotoxin bindet nicht an die GABA-Bindungsstelle, sondern wirkt möglicherweise auf Ionenkanäle und kann als Stimulans des zentralen Nervensystems exzessive Stimulation und krampfartige Reaktionen hervorrufen[2]. Picrotoxin ist ein nicht-kompetitiver Antagonist von Glycin-Rezeptoren[3].
In vitro beschleunigte die Behandlung von SCN-Zellen mit Picrotoxin(25-1000 µM) konzentrationsabhängig den zirkadianen Rhythmus der Zellaktivität. Und Picrotoxin (100 µM) beschleunigte selektiv die Akkumulation des PER-2-Proteins um etwa 3.7 Stunden, ohne dessen Stabilität zu beeinträchtigen[4]. Die Behandlung von HEK293-Zellen mit Picrotoxin(100 µM) hemmte erheblich das Expressionsniveau von 5-HT3A-Rezeptoren. Picrotoxin(10-300 µM) hemmt den 5-HT-gesteuerten Strom in Zellen konzentrationsabhängig mit einem IC50-Wert von ca. 30 µM[5].
In vivo führte die intraperitoneale Gabe von Picrotoxin(1mg/kg) über 5 Tage bei NG2-CreERTM:EYFP-Mäusen zu einer erheblichen Erhöhung der Anzahl und Dichte von OPC-Neuronenpaaren[6]. Die intraperitoneale Gabe von Picrotoxin (2mg/kg) bei weiblichen CD-Ratten erhöhte den Acetylcholin-Gehalt im Striatum um ca. 70% und den Homovanillinsäure-Spiegel um ca. 30%[7].

Vorbereitungsmethode | SCN-Zellen wurden auf Elektroden mit 30 µm Durchmesser ausgesät und drei Wochen lang in CO₂-Puffer kultiviert. Die an der Elektrodenanordnung befestigte Kulturkammer wurde mit einer fluorierten Ethylen-Polypropylen-Membran abgedeckt und in den Aufnahmeinkubator überführt. Picrotoxin (25, 50, 100, 500 und 1,000 µM) wurde der Kultur zugegeben, und Entladungen, die meistens 4 Tage anhielten, wurden zur Rhythmusanalyse aufgezeichnet. | ||

Vorbereitungsmethode | NG2-CreERTM:EYFP-Mäuse erhielten 5 Tage lang einmal täglich intraperitoneale Tamoxifen-Injektionen. Am zehnten Tag wurde das Trinkwasser durch gesüßtes Wasser ersetzt. Am zwölften Tag wurde das gesüßte Wasser durch BrdU-haltiges Wasser ersetzt. Vom 13. bis zum 17. Tag erhielten die Mäuse täglich eine Injektion mit entweder Kochsalzlösung oder Picrotoxin (1.0 mg/kg; n = 4). Die Mäuse wurden am 75. Tag, acht Wochen nach Ende der BrdU-Exposition, euthanasiert. | ||
