Bindaritは様々なモデルで抗炎活性を示すindazolic誘導体である[1]。Bindaritはp65とp65/p50媒介のMCP-1プロモーターの活性化を特異的に阻害し、発現と核局在を増やすことでFABP4と直接的に相互作用し、よってペルオキシソーム増殖因子活性化の受容体γ（PPARγ）とリポ多糖（LPS）依存性のキナーゼシグナル伝達経路を影響する[2]。Bindaritは、Chikungunyaウイルス感染による骨量減少の予防に広く使われている[3]。
In vitro（体外）実験で、Bindarit（100µM）で48時間処理したところ、TNF-α（30ng/ml）に誘発された冠動脈平滑筋細胞（CASMC）の増殖と遊走が著しく阻害された[4]。300µM Bindaritで1時間前処理したところ、Angiotensin II、endothelin-1とTGFβが誘発したヒト腎メサンギウム細胞（HRMC）の収縮、vinculinタンパク質のリン酸化が著しく減少した[5]。
In vivo（体内）実験で、100mg/kgのBindaritを10日間、毎日二回経口投与したところ、マウスで、トリニトロベンゼンスルホン酸（TNBS）誘発性の大腸炎が著しく軽減され、白血球の浸潤が減り、MCP-1の合成が下方制御された[6]。100mg/kgのBindaritを12週間毎日腹腔内注射したところ、PC-3M-Luc2異種移植マウスモデルで、腫瘍の増殖と転移が著しく阻害され、生存率が改善された[7]。100mg/kgのBindaritを28日間、毎日皮下注射したところ、癲癇重積状態のマウスで、脳損傷が軽減され、血液脳関門の完全性が維持され、神経炎症が緩和された[8]。

調製方法 |
37°C、5% CO2の環境で、0.5mg/mlのhEGF、5mg/mlのインスリン、1mg/mlのhFGF、50mg/mlのゲンタマイシン/アムホテリシン-B、5%のウシ胎児血清（FBS）を添加した平滑筋基礎培地でCASMCを培養した。細胞を24ウェルのマイクロプレート（1.5×104個の細胞/ウェル）に播種し、24時間培養した。静止状態の誘導後、Bindarit（10、25、50、100、200と300µM）の存在する/しない環境下で、細胞をTNF-α（30ng/ml）とFBS（5%）で48時間刺激し、細胞の増殖を評価した。 |

調製方法 |
温度（21-23°C）、湿度（30-35%）と明暗周期（午前7時から午後7時）がコントロールされた飼育環境下で雌BALB/cマウス（6週齢）を飼育し、標準齧歯類飼料を投与した。2.5×105個の4T1-Luc細胞/100μl PBSを、10週齢のBALB/cマウスの乳腺脂肪パッドに同所移植した。100mg/kg Bindaritを2週間、週に5日間連続で、毎日一回経口投与した。腫瘍の増殖を生物発光（BLI）でモニタリングした。 |
