グラシリン(Gracillin)は、D. quinqueloba に含まれる天然のステロイドサポニン成分であり、抗腫瘍、抗菌、抗酸化、アポトーシス誘導、および抗炎症作用を有している。
In vitroでは、グラシリンは RKO、SW480、HCT116 細胞の生存率を著しく抑制し、IC50値はそれぞれ 3.118、2.671、5.473μM であった。また、グラシリン(0、2.5、5、または10μM、36時間)は、ヒト結腸直腸癌(CRC)細胞において、用量依存的にアポトーシスを誘導した。グラシリンは、A549細胞の増殖を、2.54μmol/LのIC50値で濃度依存的かつ時間依存的に明らかに抑制し、形態学的変化を誘発した。グラシリンは、0~15μMの範囲で、IC50値が8.3μMの胃腫瘍BGC823細胞の増殖を、またIC50値が8.9μMの胃腫瘍SGC7901細胞の増殖を、それぞれ濃度依存的に抑制した。グラシリン(0~12μM、12時間)は、BGC-823細胞において濃度依存的にTIPE2の発現を誘導した。グラシリンは、肺、結腸直腸、前立腺、咽頭、および肝臓由来の様々なヒト癌細胞株において、IC50値が約1~5μMで、ミトコンドリア機能に対して用量依存的な抑制効果を示した。
In vivoでは、グラシリン(1 または 2 mg/kg、腹腔内投与、14 日間にわたり 8 回投与)は、体重を安定に保ちながら、BALB/c ヌードマウスにおける HCT116 細胞由来の異種移植腫瘍の増殖を用量依存的に抑制した。1 mg/kgのグラシリンを投与された魚は、対照群に比べて寄生虫の数が有意に少なく、グラシリンの草魚に対する急性毒性のLD50は1.64 mg/kgであった。

調製方法 |
A549細胞を6ウェルプレートに1.5×105/ウェルの密度で播種し、12時間培養した後、一連の濃度のグラシリン(0.25、0.5、1、2、4µM)で24時間処理した。細胞をHoechst 33258染色バッファーで染色し、蛍光顕微鏡を用いてアポトーシスの形態を観察した。 |

応用 |
Hoechst33258による染色後、蛍光顕微鏡観察により、グラシリンがA549細胞のアポトーシスに形態学的変化を誘発したことが確認された。対照群と比較して、グラシリン処理群ではアポトーシス細胞に典型的な形態学的特徴が認められた。具体的には、細胞数が比較的減少しており、核はピコノシスを呈し、クロマチンは核膜の下で塊状に凝縮しており、鮮やかな青色の蛍光染色を示していた。 |
