Description of Daptomycin

Daptomycin은 빠른 박테리치다이랙티브 활동을 가진 리포펩타이드 계 항생제로 그람 양성 박테리아에 대한 효능이 있다.Daptomycin은 엔터로코커, 스타피로코커, 스트레포코커, 코르네바케리아 그리고 정체기 지속 박테리아 보렐리아 버그도르페리 [2]에 대한 체외 활동이 있다. Daptomycin의 항균 메커니즘은 대부분의 항생제와 다른다. 아실글리세롤 인산염을 이용하여 세포막에 삽입되어 뭉치고 세포막의 곡률을 변화시키고 천공을 냄으로써 세포의 극성을 유발하고 세포 괴사로 이어진다 [3].

체외 실험에서 Daptomycin (5μg/ml)은 30분 내에 황색 포도 상구균 세포 생존력을 99% 이상, 멤브레인 잠재력 90% 이상 감소시킨다[4]. Daptomycin (100 μg/ml)은 고밀도 지수성 성장하는 황색 포도 상구 균 ATCC 29213 세포에 60분 동안 처리되어 매우 빠른 박테리치다이렉트 효과를 보여 박테리아 수를 3 로그로 줄였다[5].

체내 실험에서 Daptomycin (20 mg/kg; i.p.)은 에테르로코커로 접종한 복막염 쥐의 생존율을 현저히 향상시키고 치료 후 6시간 내에 살아있는 세균의 수와 호중구 화학 중성 백혈구 순화인자 KC 수준을 줄였다[6]. Daptomycin (60 mg/kg; s.c.)은 Sprague-Dawley CD 쥐의 황금균 골수염 치료에서 중간 정도의 효능을 가지고 있다[7].
