Morusin은 Morus alba (Moraceae) 뿌리 피질에서 발견되는 중요한 프레닐화된 플라보노이드로 전립선암 치료에서 상당한 항종양 활성을 보인다. 프레닐화된 플라보노이드는 플라보노이드 골격과 지용성 프레닐 측쇄가 결합된 독특한 플라보노이드 클래스로 세포독성, 항염증, 알츠하이머병 예방, 당뇨병 예방 등 기타 효과를 포함한 큰 잠재적 생물학적 활성을 가지고 있다[2]. Morusin은 염증성 폐질환의 새로운 효과적인 점액 조절제 후보이다[3].
체외 실험에서 Morusin(0-10μM)으로 48시간 처리한 PC3 세포는 시간 및 농도 의존적으로 현저하게 세포 생존율을 저해하고 염증성 사이토카인 수준을 감소시켰다[4]. 24시간 동안 혈청을 제거한 NCI-H292 세포에서 Morusin(1-100μM)을 30분 동안 투여하면 점액 5AC(MUC5AC)의 생성이 현저하게 억제되어 과도한 점액 분비로 인한 다양한 폐 염증 반응을 조절할 수 있다[3]. Morusin(1-5mg/L)으로 72시간 처리한 암 세포주(MKN45 및 SGC7901)에서 Morusin은 암세포의 성장과 증식을 효과적으로 억제했다[5].
체내 실험에서 Morusin(5mg/kg/5일)을 28일 동안 위내 투여한 간세포절제된 쥐에서 Morusin은 에스트로겐 결핍 유도 골다공증을 완화하고 대퇴골 조직에서 RUNX2와 β-카테닌의 표현을 감소시켰다[6]. 내측 반월상 연골 불안정성 골관절염(DMM) 쥐 모델에서 Morusin(5mg/kg/2일)을 56일 동안 위내 투여하면 Morusin은 연골의 침식과 파괴를 완화하며 관련 염증 인자와 단백질의 발현 수준을 감소시킵니다[7].

제조 방법 | 96웰 배양판에서 PC3 세포를 4×10⁴ 세포/웰의 밀도로 심었습니다. 약물 처리 전에 세포가 판에 부착할 수 있도록 24시간 동안 배양했습니다. Morusin(0, 1.25, 2.5, 5, 10µM)의 연쇄 희석액을 세포에 적용해서 반 억제 농도 50을 측정했습니다. 48시간의 처리 기간 후 4℃에서 1시간 동안 10% 트리클로로아세트산을 적용해서 세포를 고정시켰습니다. 그리고 0.4%의 Sulforhodamine-B를 30분 동안 처리한 후 1% 아세트산으로 헹구고 공기 중에서 건조시켰습니다. 염료는 pH 10.5의 10mM 트리스 기지 용액으로 판을 헹구어 제거되었습니다. 이 후에 ELISA 마이크로플레이트 리더를 사용해서 570nm 파장에서 웰의 광학 밀도를 정량화했습니다. |
