FT671 ist ein potenter und selektiver Inhibitor der Ubiquitin-sprzifischen Protease 7 (USP7) mit einem IC50 Wert von 52nM[1]. USP7 kann MDM2 deubiquitinieren, was zu einer Abnahme des intrazellulären p53[2]-Spiegels führt. FT671 besitzt Antitumoraktivität[3].
In-vitro-Studien zeigten, dass eine Vorbehandlung von A549-Zellen mit FT671 (0-24µM) über 4 Stunden den Abbau der intrazellulären SP1-and β-Catenin Proteine signifikant förderte[4]. Die Behandlung von HCT116-Dickdarmkrebszellen mit FT671 (10µM) über 5 Minuten schwächte die intrazelluläre Proteinubiquitinierung unter Bedingungen des USP7- Knockdown signifikant ab[5].
In vivo-Studien zeigten, dass die orale Behandlung von MM.1S-Zell-Xenotransplantat-Mäuden mit FT671 (100, 200mg/kg) das Tumorwachstum dosisabhängig signifikant hemmte und gut verträglich war[6].

Vorbereitungsmethode | 6 bis 8 Wochen alte weibliche NOD-SCID-Mäuse wurde 24 Stunden vorder sekutanen Inokulation von 5×106 MM.1S-Tumorzellen in einer 1:1 Mischung aus RPMI-1640 und Matrigel mit einer Co60-Bestrahlungsquelle(2Gγ) bestrahlt. Die Wirkamkeitsstudie wurde initiiert, als die Tumoren ein durchschnittliches Volumen von 159 mm3 erreicht hatten. Tumortragende Mäuse wurden nach dem Zufallsprinzip den Behandlungsgruppen zugewiesen, so dass jede Behandlungsgruppe dasselbe durchschnittliche Tumorvolumen hatte. Mäuse erhielte eine Einleitungsdosis von 25mg/kg FT671 24 Stunden vor Beginn des Dosierungsschemas von 100 oder 200mg/kg FT671 (einmal täglich, per os durch porale Sondierung). 10% DMA/90% PEG400 diente als Vehikelkontrolle.Für die statistische Analyse der Unterschiede im Tumorvolumen zwischen den Behandlungsgruppen wurde eine 2-Wege-ANOVA mit wiederholten Messungen durchgeführt, gefolgt von einer Korrektur für multiple Vergleiche unter Verwendung statistischer Hypothesentests |

Anwendungen | Die Behandlung von Mäusen mit FT671 führte zu einer signifikanten dosisabhängigen Tumorwachstumshemmung. FT671 wurde selbst bei hohen Dosen gut vertragen, und während der Studie wurde kein signifikanter Gewichtsverlust oder Kachexie beobachtet. |
