MG 149 ist ein Inhibitor von Histon-Acetyltransferasen(HAT) mit IC50-Werten von 74 μM für Tip60 und 47 μM für MOF.
In vitro verhinderte die Behandlung mit 2 μM MG 149 über 12 Stunden die H4K16-Acetylierung. Sie betraf serumentzogene A549- und U2OS-Zellen nach DNA-Schädigung durch Doxorubicin[2]. Die Behandlung mit 100 μM MG 149 über 30 Minuten verhinderte erheblich die UV-induzierte XPF-Acetylierung in HeLa-Zellen[3]. Im Dosisbereich von 0-30 μM hemmte MG 149 nach 20 Stunden die katalytische Aktivität von TIP60 in Wildtyp-2D10-Zellen dosisabhängig. Zudem kehrte es den latenten HIV-Zustand um[4].
In vivo linderte 1mg/kg MG 149(intraperitoneal verabreicht) die HDM-induzierte Atemwegshyperreaktivität(AHR) und Atemwegsentzündung in murinen allergischen Asthmamodellen[5]. MG 149 wurde 14 Tage lang in einer Dosis von 1.6mg/kg intraperitoneal injiziert. Dadurch blockierte es die erhöhte Glutamat-Freisetzung im ventralen hippocampalen CA1(vCA1) nach elektrischer Stimulation des dorsomedialen präfrontalen Kortex(dmPFC) bei CRS-Mäusen[6].

Vorbereitungsmethode | Die Zellen wurden 48 Stunden lang in einem Medium mit 0.5% Serum kultiviert. Nach einer 10-stündigen Behandlung mit 2 μM MG 149 und 10 μM C646 wurde 10 μM Doxorubicin zugegeben, um DNA-Schäden zu induzieren und der Acetylierungsgrad von H4K16 wurde nach 2 Stunden gemessen. |

Anwendungen | Der Inhibitor MG 149, der auf Tip60 abzielt, verhinderte die H4K16-Acetylierung als Reaktion auf durch Doxorubicin induzierte DNA-Schäden. Diese Acetylierung wurde jedoch nicht durch C646 beeinflusst, das p300 hemmt, und zwar in serumdeprivierten A549- und U2OS-Zellen. |

Vorbereitungsmethode | Mäusen wurde über zwei Wochen an 5 aufeinanderfolgenden Tagen intranasal 35ml (0.7mg/ml PBS) Gesamtproteinextrakt aus HDM(Hausstaubmilben) verabreicht, um ein allergisches Asthma-Modell zu erzeugen. In der Gruppe HDM+MG 149 erhielten die narkotisierten Mäusen 60 Minuten vor der HDM-Provokation eine intraperitoneale Injektion von MG 149 (1mg/kg). Kontrolltiere erhielten lediglich PBS. |
