TCEP hydrochloride ist ein nicht-thiolhaltiges Reduktionsmittel. TCEP hydrochloride assoziiert sich nicht mit Hg2+[1]. TCEP hydrochloride fördert die Erzeugung von SOD1-Monomeren. Dadurch verhindert TCEP hydrochloride den NO-induzierten Anstieg der Dismutase-Aktivität und die Abnahme von ROS[2].
In-vitro-Wirksamkeitstest zeigten: TCEP hydrochloride verstärkte die NF-kappaB-DNA-Bindung dosisabhängig im Konzentrationsbereich von 0.25 bis 6mM[1].
Ein in-vitro-Versuch ergab: Eine Behandlung mit 100μM TCEP hydrochloride erhöhte die Qualität und Entwicklungsfähigkeit von in vitro befruchteten Embryonen. TCEP hydrochloride senkte den oxidativen Stress in Schweine-Oozyten[3]. In der SH-SY5Y-Zelllinie bot 1mM TCEP hydrochloride den maximalen Schutz vor der Hemmung der [(3)H]-NA-Freigabe durch BoNT/B und war nicht toxisch[4]. TCEP hydrochloride (0.01mM) entfernte kein Fe3+ aus Tf. TCEP hydrochloride schützt Thiolgruppen für die Kopplung mit Maleimid. Außerdem stört TCEP hydrochloride die Maleimid-Kopplung selbst nicht[5]. In vitro wurden menschliche hepatozelluläre(HepG2) Zellen mit 100, 200 und 400μM TCEP hydrochloride über 3 Tage behandelt. Der oxidative Stress, die Esterase-Aktivität, der Ca2+-Einstrom und die Dysfunktion von ΔΨm nahmen zu. Bei den mit 400μM behandelten Zellen lag der subG1-Apoptosepeak bei 65.96%[6].
In vivo erhielten erwachsene ICR-Mäuse oral 20mg/kg und 60mg/kg TCEP hydrochloride über 9 Wochen. TCEP erhöhte die Gewichtszunahme, milderte die Hypertriglyceridämie und die hepatische Steatose. Das ging mit einer Hochregulation des Expressionsniveaus leberspezifischer Lipogenese-Gene einher. TCEP hydrochloride veränderte zudem die Konzentrationen einiger Lebermetabolite[7].

Insgesamt 30 erwachsene Kunming-Mäuse wurden zufällig aufgeteilt: Normale Kontrollgruppe(0mg/kg·Tag), Niedrigdosis-Gruppe TCEP hydrochloride(10mg/kg·Tag) und Hochdosisgruppe TCEP hydrochloride(100mg/kg·Tag). Die Gabe erfolgte kontinuierlich durch orale Sondierung über 30 Tage. |

Im Vergleich zur Kontrollgruppe sank die Wasseraufnahme in der Hochdosisgruppe erheblich. Der Organindex von Leber und Milz stieg merklich an. Im Wasserlabyrinth-Test war die Fluchtlatenz bei den TCEP-behandelten Mäusen länger als bei der Kontrollgruppe. Bei der Hochdosisgruppe wurde die gesamte Schwimmstrecke erhöht. Die Schwimmzeit im Zielquadranten war gegenüber der Kontrollgruppe erheblich verkürzt. |
