77PD-Q is a para-phenylenediamine quinone, which prevalently exists in the airborne particulates [1]. 77PD-Q induces endothelial-to-mesenchymal transition (EndMT) of cerebral microvascular endothelial cells by upregulating PAI-1, resulting in blood-brain barrier (BBB) dysfunction[2]. 77PD-Q has been widely used as a model compound to investigate the toxicity to aquatic organisms in sediment samples from urban rivers, coasts and deep-sea areas[3]. 77PD-Q has been widely used as an internal reference to optimize the conditions for ion chromatography and mass spectrometry analysis, thereby facilitating the identification and separation of related compounds respirable fine particulates[4].
In vitro, 77PD-Q (1μM) treatment for 24 hours significantly stimulated the release of inflammatory cytokines and lactate dehydrogenase (LDH) in hCMEC-D3 cells and promoted cell apoptosis[2].

Cell lines | hCMEC-D3 cells |
Preparation Method | hCMEC-D3 cells were grown in endothelial cell medium with 5% (v/v) fetal bovine serum (FBS), 1% endothelial cell growth supplement, and 1% penicillin/streptomycin solution in the presence of 5% CO2 at 37°C. Cells (5×103 cells/ml) were seeded in 96-well plates and allowed to adhere in a 5% CO2 incubator at 37°C overnight. Cells were treated with different concentrations of 77PD-Q (0, 0.01, 0.10, 1.00μM) for 24h. The cell viability was evaluated. |
Reaction Conditions | 0, 0.01, 0.10, 1.00μM; 24h |
Applications | 77PD-Q treatment significantly reduced cell viability of hCMEC-D3 cells in a dose-dependent manner. |

Method for preparing DMSO master liquid: mg drug pre-dissolved in μL DMSO ( Master liquid concentration mg/mL, Please contact us first if the concentration exceeds the DMSO solubility of the batch of drug. )
Method for preparing in vivo formulation: Take μL DMSO master liquid, next addμL PEG300, mix and clarify, next addμL Tween 80, mix and clarify, next add μL saline, mix and clarify.
Method for preparing in vivo formulation: Take μL DMSO master liquid, next add μL Corn oil, mix and clarify.
Note: 1. Please make sure the liquid is clear before adding the next solvent.
2. Be sure to add the solvent(s) in order. You must ensure that the solution obtained, in the previous addition, is a clear solution before proceeding to add the next solvent. Physical methods such as vortex, ultrasound or hot water bath can be used to aid dissolving.
