MSA-2 ist ein oral erhältlicher Nicht-Nukleotid-Interferon-Genstimulator(STING)-Agonist, mit EC50-Werten von 8,3μM und 24μM für Human-STING-Subtypen WT bzw. HAQ[1].MSA-2 weist eine höhere Wirksamkeit der sauren Tumormikroumgebung auf, wo das kleine Molkül eine nichtkovalente Dimerisierung zur Bildung eines bioaktiven Liganden erfährt[2]. MSA-2 in Lösung existiert als Monomere und nichtkovalente Dimere in einem Gleichgewicht, das eine starke Tendenz zum monomeren Zustand hält[3].
In vitro inhibitierte MSA-2 (20-50 μM) Behandlung von PK-15 -Zellen, die mit Seneca-Talvirus (SVV) infiziert sind, die SVV-Replikation in den Zellen, den STING-Signalweg und induzierte die Expression von Zytokinen IFN-β, IL-6, und TNF-α[4]. Die Behandlung mit MSA-2 (10, 50 μM) von RAW264,7-Zellen erhöhte intrazellulären IFN-β-Spiegeln[5]. Die Behandlung mit MSA-2 (25μM) humaner Monozyten für 20 Stunden eliminierte die LPS-induzierte IL-10 und IL-19 Produktion, während IL-1β und TNF-α nicht inhibitiert wurden[6].
In vivo hemmte die Behandlung von MSA-2 (p.o. 60 mg/kg or s.c. 50 mg/kg) bei Mäusen mit Kolonkrebsmodellen stark das Tumorwachstum und erhöhte Spiegel an Zytokinen IFN-β, IL-6, und TNF-α in Tumorzellen[1]. MSA-2 (150 μg) wurde intrathekal verabreicht wodurch es die Mäuse mit Kolonkrebs- und Melanomamodellen, verbesserte die Überlebensrate und verstärkte die Infiltration und Aktivität von zytotoxischen T-Zellen in Tumoren und trugen zur Tumorregression bei[5].

Vorbereitungsmethode | MC38-Zellen wurden subkutan in 6 Wochen alte weibliche C57BL/6 Mäuse injiziert, um Tumormodelle zu etabilieren. Wenn die Tumorvolumen etwa 100mm3 erreichten, wurden die Mäuse zufällig in 3 Gruppen unterteilt : Kontrolle, MSA-2, and MSA-2-Pt. Die Mäuse waren unter Kontrolle injiziert, 150μg MSA-2, und 150μg MSA-2-Pt für drei Dosen. |
