Fc receptors (FcRs) are expressed on various cell types, including monocytes, granulocytes, B cells, and dendritic cells. Cells expressing FcRs may occasionally yield false-positive or false-negative results during immunofluorescence staining due to the FcR-mediated binding of immunoglobulin (Ig) Fc regions.
The Human Fc Receptor Blocking Solution has been specifically developed to block non-specific staining caused by FcRs without interfering with antibody-mediated specific staining of human cells. This product is compatible with flow cytometry staining protocols utilizing anti-human CD16 (Clone 3G8), CD32 (Clone FUN-2), and CD64 (Clone 10.1) antibodies.
It is recommended to add 5 µL of the Fc Receptor Blocking Solution per 1 million cells in a total staining volume of 100 µL. After mixing, incubate the sample at room temperature for 5–10 minutes, and then proceed with staining using the antibodies of interest. No washing step is required between the blocking and immunostaining procedures.
