Docosahexaenoic Acid ethyl ester is an ethyl ester derivative of an Omega-3 polyunsaturated fatty acid. Docosahexaenoic acid ethyl ester enhances 6-hydroxydopamine-induced neuronal damage by inducing lipid peroxidation in the mouse striatum. Docosahexaenoic acid ethyl ester can be used in research related to neurodegenerative diseases[1-4].
In vitro, Docosahexaenoic Acid ethyl ester (10, 30, 50μg/ml) was incubated with human squamous cell carcinoma HSC-4 cells for 15 hours. Docosahexaenoic Acid ethyl ester significantly inhibited the proliferation of HSC-4 cells[5]. Docosahexaenoic Acid ethyl ester (400μg/ml) was used to treat 95D non-small cell lung cancer cells for 24 hours. Docosahexaenoic Acid ethyl ester slightly reduced cell viability and significantly decreased the expression of NF-κB protein[6].
In vivo, Docosahexaenoic Acid ethyl ester (200mg/kg; once daily) was orally administered to male Mongolian gerbils for 10 consecutive weeks, followed by 10 minutes of bilateral common carotid artery occlusion. Docosahexaenoic Acid ethyl ester significantly inhibited the increase in hydroxyl radical production and the elevation of the lipid peroxidation product malondialdehyde caused by cerebral ischemia-reperfusion, prevented the decrease in reduced glutathione levels and the activities of glutathione peroxidase and catalase, and attenuated hyperlocomotion and neuronal loss in the hippocampal CA1 region[7]. Docosahexaenoic Acid ethyl ester (200mg/kg; once daily) was orally administered to Mongolian gerbils for 10 consecutive weeks, followed by 10 minutes of bilateral common carotid artery occlusion. Docosahexaenoic Acid ethyl ester significantly reduced the production of eicosanoids after cerebral ischemia-reperfusion, prevented the impairment of regional cerebral blood flow, and alleviated cerebral edema[8].

| Cell experiment [1]: | |
Cell lines | 95D non-small-cell lung cancer cells |
Preparation Method | 95D cells were cultured in RPMI 1640 medium containing 10% fetal bovine serum and 0.1% penicillin-streptomycin at 37°C with 5% CO₂. 95D cells were treated with Docosahexaenoic Acid ethyl ester at 400μg/ml for 24 hours. |
Reaction Conditions | 400μg/ml; 24h |
Applications | Docosahexaenoic Acid ethyl ester had a significant effect on the cell viability of 95D cells. Docosahexaenoic Acid ethyl ester had no significant effect on cell apoptosis, but significantly decreased NF-κB protein expression. |
| Animal experiment [2]: | |
Animal models | Male Mongolian gerbils (21-day-old weanling) |
Preparation Method | Gerbils were orally pretreated with Docosahexaenoic Acid ethyl ester emulsified in 5% gum Arabic solution, once a day, for 10 weeks, and then subjected to transient forebrain ischemia by bilateral common carotid occlusion for 10 minutes followed by reperfusion. |
Dosage form | 200mg/kg; oral; once a day for 10 weeks |
