Cyanine5.5 maleimide is a near-infrared fluorescent dye labeling reagent (Ex=673nm; Em=707nm). The maleimide group reacts with the SH-thiol group to form a thioether bond, thereby labeling proteins, peptides, antibodies, etc., containing cysteine molecules.
This plan is based on literature, please modify it to meet your specific needs.
1. Cyanine5.5 maleimide stock solution: Dissolve Cyanine5.5 maleimide dye in anhydrous DMSO to prepare a 10mM stock solution. Dissolve thoroughly by pipetting or vortexing.
Note: Long-term storage of the dye stock solution may reduce the activity of the dye; it should be used as soon as possible. It is recommended to aliquot the Cyanine5.5 maleimide stock solution and store it at -20℃ or -80℃ protected from light, avoiding repeated freeze-thaw cycles.
2. Protein stock solution: Dilute the protein to be labeled to 2mg/ml with PBS/HEPES, pH=7.5±0.5.
Note: (1) If the protein concentration is below 2mg/ml, the binding efficiency will be significantly reduced. For optimal labeling efficiency, a final protein concentration range of 2-10mg/ml is recommended. (2) The pH of the protein solution should be 7.5±0.5. Too low a pH will affect the labeling efficiency, while too high a pH may lead to non-specific labeling. (3) If your protein does not contain free cysteine, you need to treat the protein with a thiol reducing agent such as DTT or TCEP to generate thiol groups. Before coupling, remove free DTT from the protein solution by dialysis or gel filtration.
3. Calculate the volume of Cyanine5.5 maleimide and the protein to be labeled at a molar ratio of 5:1. Slowly add the Cyanine5.5 maleimide dye to the protein sample, gently shake to mix, and briefly centrifuge to collect the sample at the bottom of the reaction tube. Avoid vigorous mixing to prevent denaturation and inactivation of the protein sample.
Note: The amount of dye required for the labeling reaction depends on the protein properties. It is recommended to experiment within a molar ratio range of 2-10:1.
4. Place the reaction tube in a dark place and incubate gently at room temperature for 30-60 minutes.
5. Wash away the free dye using a desalting centrifuge column and PBS, and collect the protein conjugate.
